Cyclic fluctuations of estradiol and progesterone in females influence neuronal activity in the ventrolateral division of the ventromedial hypothalamic nucleus (VMNvl), through the activation of progesterone receptors (PRs) and estrogen receptors (ERs). The expression of ER and PR in the VMNvl is influenced by their cognate ligands and is a central upstream trigger in the pathway of VMNvl-dependent modulation of endocrine responses. By studying the role played by estradiol and progesterone in PR and ERa expression in the VMNvl along the estrous cycle and how the two receptors interact in the same neuron, we aim to evaluate the synergistic action of both ovarian hormones in the regulation of VMNvl activity. In animals at all phases of the estrous cycle, the number of VMN neurons expressing PR or ERa was estimated by stereological methods, and the percentage, and rostro-caudal distribution, of neurons simultaneously expressing both receptors was determined. The highest number of PR-immunoreactive neurons was seen at proestrus, and of ERa-immunoreactive neurons was seen at proestrus and metestrus. The ERa/PR co-localization is increased at caudal levels. Approximately half the neurons expressing PR co-express ERa, a proportion that stays constant along the estrous cycle. The percentage of ERa neurons co-expressing PR changes from 60% at proestrus to 40% at metestrus. Fluctuations in circulating ovarian hormone levels promote coordinated changes in PR and ERa expression and co-localization. This may be an important mechanism in the regulation of input relayed by the VMNvl, allowing a precise modulation of endocrine responses.

Reconsumption of ethanol after withdrawal is a hallmark for relapse in recovering patients with alcohol use disorders. We show that the preference of Drosophila melanogaster to reconsume ethanol after abstinence shares mechanistic similarities to human behavior by feeding the antirelapse drug acamprosate to flies and reducing the ethanol consumption preference. The Drosophila cellular stress mutant hangover also reduced ethanol consumption preference. Together with the observation that an increasing number of candidate genes identified in a genome-wide association study on alcohol use disorders are involved in the regulation of cellular stress, the results suggest that cellular stress mechanisms might regulate the level of ethanol reconsumption after abstinence. To address this, we analyzed mutants of candidate genes involved in the regulation of cellular stress for their ethanol consumption level after abstinence and cellular stress response to free radicals. Since hangover encodes a nuclear RNA-binding protein that regulates transcript levels, we analyzed the interactions of candidate genes on transcript and protein level. The behavioral analysis of the mutants, the analysis of transcript levels, and protein interactions suggested that at least two mechanisms regulate ethanol consumption preference after abstinence—a nuclear estrogen-related receptor-hangover-dependent complex and peroxisomal trans-2-enoyl-CoA reductase (dPECR)-dependent component in peroxisomes. The loss of estrogen-like receptor and dPECR in neurons share a protective function against oxidative stress, suggesting that the neuroprotective function of genes might be a predictor for genes involved in the regulation of ethanol reconsumption after abstinence.

Este é um estudo quantitativo realizado entre 2017 e 2018 em uma universidade pública estadual da Bahia, com o objetivo de avaliar a prática de binge drinking em universitários. Utilizou-se questionário autoaplicável e análise descritiva e inferencial. Participaram 150 universitários, sendo 52% mulheres e 48% homens que afirmaram ingerir bebidas alcoólicas, com média de idade de 23,2 anos (dp±3,6). Houve associação entre beber para amenizar a ansiedade e a prática do binge drinking (p=0,01); no uso moderado do álcool relatou-se não sentir nada ou sentir sonolência após o consumo da bebida, enquanto os que faziam uso nocivo, a sensação era de alegria (p<0,0001). No padrão de consumo típico do binge drinking, 41% eram mulheres e 59% eram homens. As diferenças estatisticamente significantes foram: quanto ao sexo masculino – ter sofrido ou causado prejuízos a terceiros (p=0,04); quanto ao sexo feminino – possuir benefício social (p=0,0001) e esconder dos pais que faziam uso excessivo do álcool (p=0,01). Os fatores de risco observados: sexo, início precoce do uso do álcool, ingresso na universidade e influência dos pares. Já os fatores de proteção: estar casado e ter religião. A universidade precisa entender seu papel nesse cenário incluindo atividades culturais e ofertando acesso a conhecimento sobre riscos e a redução do consumo de alcool nos universitários.
