This study investigated the diversity and abundance of bacterial lacasse-like genes in different particle size fractions, namely sand, silt, and clay of sediments in a subtropical mangrove ecosystem. Moreover, the effects of nutrient conditions on bacterial laccase-like communities as well as the correlation between nutrients and, both the abundance and diversity indices of laccase-like bacteria in particle size fractions were also studied. Compared to bulk sediments, Bacteroidetes, Caldithrix, Cyanobacteria and Chloroflexi were dominated in all 3 particle-size fractions of intertidal sediment (IZ), but Actinobacteria and Firmicutes were lost after the fractionation procedures used. The diversity index of IZ fractions decreased in the order of bulk > clay > silt > sand. In fractions of mangrove forest sediment (MG), Verrucomicrobia was found in silt, and both Actinobacteria and Bacteroidetes appeared in clay, but no new species were found in sand. The declining order of diversity index in MG fractions was clay > silt > sand > bulk. Furthermore, the abundance of lacasse-like bacteria varied with different particle-size fractions significantly (p < 0.05), and decreased in the order of sand > clay > silt in both IZ and MG fractions. Additionally, nutrient availability was found to significantly affect the diversity and community structure of laccase-like bacteria (p < 0.05), while the total organic carbon contents were positively related to the abundance of bacterial laccase-like genes in particle size fractions (p < 0.05). Therefore, this study further provides evidence that bacterial laccase plays a vital role in turnover of sediment organic matter and cycling of nutrients.

Diversidade de Pseudomonas fluorescentes em diferentes sistemas de manejo do solo e rotação de culturas RES UMOO objetivo neste trabalho foi avaliar o efeito do sistema de manejo do solo (plantio direto e plantio convencional) e da rotação de culturas na diversidade de Pseudomonas fluorescentes isoladas de rizoplano e no solo das entrelinhas de trigo, com base nas características morfológicas e genotípicas destas bactérias. Na análise morfológica, a maioria dos isolados obtidos de rizoplano apresentou características de P. fluorescens, enquanto naqueles obtidos de solo foram observadas características de P. putida. A análise de correspondência, realizada com os dados morfológicos, comprovou que os isolados de solo são mais semelhantes aos da mata que os de rizoplano. A população de Pseudomonas do rizoplano é muito homogênea e não sofreu efeito do sistema de manejo do solo nem da rotação de culturas. Por outro lado, a população do solo das entrelinhas formou dois grupos, um para o plantio direto e outro para o convencional. A análise por ARDRA (Amplified Ribosomal DNA Restriction Analysis) resultou em três grupos genotípicos, com predominância dos isolados obtidos de rizoplano, com características morfológicas de P. fluorescens. Esses dados indicam a existência de um processo de seleção da comunidade bacteriana, controlado pela planta, que apresenta em seu rizoplano uma prevalência Pseudomonas com características morfológicas de P. fluorescens. Palavras-chave: análise multivariada, características morfológicas, marcador molecularFluorescent Pseudomonas diversity in different soil management and crop rotation systems AB S TR ACTThe objective of this work was to evaluate the effects of different soil management (no-tillage and conventional system) and crop rotation systems on the diversity of fluorescent Pseudomonas obtained from the rhizoplane and soil samples of wheat interlines, assessed by morphological and genotypic data. Morphological data analysis indicates that the majority of the isolates from the rhizoplane show characteristics of P. fluorescens, while the isolates from the soil have characteristics of P. putida. The correspondence analysis based on morphological data showed that the soil isolates are more similar to the ones of a forest area than the ones of the rhizoplane. Pseudomonas population from the rhizoplane is very homogeneous, without any effect of both management system and crop rotation. On the other hand, population from the interline soil samples formed two groups, one for the no-tillage system and another for the conventional system. ARDRA analysis showed three genotypic groups, dominated by the isolates obtained from the rhizoplane, with morphological characteristics of P. fluorescens. These data indicate a selection process on the bacterial community controlled by the plant, where the presence of fluorescent Pseudomonas with morphological characteristics of P. fluorescens in the wheat rhizoplane is prevalent.
