Changes in H2O2 content and antioxidant enzyme gene expression during the somatic embryogenesis of Larix leptolepis

Abstract: Hydrogen peroxide (H 2 O 2 ) content and transcript levels of genes encoding superoxide dismutase (SOD), catalase (CAT), and ascorbate peroxidase (APX) antioxidant enzymes were investigated during different stages of somatic embryogenesis in Larix leptolepis. H 2 O 2 content was lowest on day 0 when embryogenic callus was incubated on Murashige and Skoog (MS) medium supplemented with polyethylene glycol (PEG). This content began to increase when callus was transferred to the same medium but containing abscisic… Show more

“…These high ROS doses can damage DNA, proteins and carbohydrates, resulting in cell death (Mittler et al 2004). To cope with different internal and external stresses, plants have developed several antioxidant enzymes to detoxify ROS and protect their cells from oxidative injury (Mittler et al 2004;Chatzissavvidis et al 2008;Sun et al 2010;Zhang et al 2010;Lokhande et al 2011). Catalase belongs to an important detoxifying system, the physiological role of catalase is to break down H 2 O 2 in the cell (del Rio et al 2002;Scandalios 2002).…”

“…)-ABA is able to stimulate embryo maturation upon rapid conversion to phaseic and dehydro-phaseic acid (Dunstan et al 1992;Stasolla et al 2002). Zhang et al (2010) recently reported the role of exogenously applied ABA on H 2 O 2 content during different stages of embryo development, further suggesting that the promotion of somatic embryos is ABA-dependent. For many plants, ABA does not promote somatic embryo maturation when added alone to the culture medium; however, it has been found to be …”

“…In a similar way, the osmotically inducible protein OsmC (s155) was also identified in LI, and it is involved in stress responses, particularly in defence against oxidative stress caused by exposure to organic hydroperoxides (Lesniak et al 2003). ROS are important signalling molecules reported to act in plant responses to biotic and abiotic stresses (Delledone et al 2001) and in the development of SE of conifers (Zhang et al 2010). The generation of hydrogen peroxide and its regulation by the activation of antioxidant enzymes have been recorded at the beginning of SE formation (Lilik et al 2005;Ma et al 2012a, b).…”
