Cepharanthine inhibits hepatocellular carcinoma cell growth and proliferation by regulating amino acid metabolism and suppresses tumorigenesis in vivo

Abstract: Cepharanthine (CEP), a natural compound extracted from Stephania cepharantha Hayata, has been found to have the potential to treat a variety of tumors in recent years. This study aims to evaluate the anti-hepatocellular carcinoma (HCC) effect of CEP and determine its in-depth mechanism. In this study, Hep3B and HCCLM3 cells were selected to evaluate the antitumor effects of CEP in vitro, whereas tumor xenograft in nude mice was performed to make in vivo anti-tumor assessment. RNA-sequence (RNA-seq) was used to… Show more

“…Microbial secondary metabolites, such as alkaloids, phenols, antibiotics, and pigments, determine the specificity and function of the flora. Tumors have various in vivo metabolisms, including amino acid metabolism associated with liver cancer cells ( Feng et al, 2021 ), Glucose metabolism is associated with gastric cancer ( Fu et al, 2022 ). Lipid metabolism is related to liver cancer ( Pan et al, 2021 ) and glycolysis ( Li et al, 2022 ; Pan et al, 2022 ).…”

“…In addition, CEP also suppressed endogenous ANO1 currents, significantly inhibited cell proliferation and migration, and induced death in lung cancer cells [ 104 ]. In addition, CEP could be used as a candidate drug for HCC that suppresses cell migration and proliferation [ 105 ]. IR-783, a near-infrared heptamethine cyanine dye, has a potential anti-cancer role.…”

“…Moreover, perturbation of amino acid metabolism was also correlated with the progression of hepatic live diseases ( 51 ). Noticeably, cepharanthine treatment could inhibit HCC cell proliferation and migration by regulating amino acid metabolism ( 52 ). Of note, hepatic tissue in individuals stores almost 70% of retinoids ( 53 ); as reported, the inhibition of retinoids or the loss of hepatic retinoid signaling potentially leads to oxidative stress ( 54 ), which was associated with the progression of liver diseases ( 55 ).…”
