Centrifugal microfluidics for cell analysis

Abstract: Over the past two decades, centrifugal microfluidic systems have successfully demonstrated their capability for robust, high-performance liquid handling to enable modular, multi-purpose lab-on-a-chip platforms for a wide range of life-science applications. Beyond the handling of homogeneous liquids, the unique, rotationally controlled centrifugal actuation has proven to be specifically advantageous for performing cell and particle handling and assays. In this review we discuss technologies to implement two imp… Show more

“…[10][11][12][13] Centrifugal microfluidics often utilize intrinsic, non-contact pumping and valving mechanisms which are ideal for liquid handling. [14][15][16] However, the handling of liquid/vapor PCR samples in microfluidic devices is not a simple task when confronted with the high temperatures and pressures created within the PCR chamber region.…”

“…This is the so-called "lab-on-a-disc" strategy. 14,15 In an endeavor to develop a compact, user-friendly, and cost-efficient sample-to-answer device for highly accurate target cell counts, we introduce here a novel method for centrifugo-magnetophoretic cell separation that is amenable to complete integration into a lab-on-a-disc platform. 14,16 As a proof-of-concept and an intermediate step, this work implements a bead-based immunomagnetic isolation of cells expressing the CD4 antigen on the cell surface-CD4-expressing cells being the primary host cell of the HIV virus and hence of prime diagnostic relevance.…”
