Cellular turnover and extracellular matrix remodeling in female reproductive tissues: functions of metalloproteinases and their inhibitors

Abstract: Female reproductive tissues possess a unique ability to accommodate a remarkable amount of cell turnover and extracellular matrix (ECM) remodeling following puberty. Cellular structures within ovary, uterus, and mammary tissue not only change cyclically in response to ovarian hormones but also undergo differentiation during pregnancy, and eventually revert to that resembling the pre-pregnant stage. Cell proliferation, apoptosis, invasion, and differentiation are integral cellular processes that are precisely r… Show more

“…Accumulating experimental evidence indicates the MMPs are important mediators of the ovulatory process (Brannstrom et al 1988, Butler et al 1991. The relative abundance of MMPs vs their endogenous proteinase inhibitors (TIMPs) dictates net ECM deposition vs degradation (Fata et al 2000). Results of the present studies demonstrated that mRNA and protein for TIMP-3 and TIMP-4 were increased in bovine preovulatory follicles following the gonadotropin surge, suggestive of a potential role for both TIMPs in regulation of preovulatory MMP activity.…”

“…Since ovulation is a prerequisite for fertilization and embryonic development, the mechanism of ovulation has been a subject of investigation for some time. Experimental evidence supports the theory that proteolytic degradation of the extracellular matrix (ECM) at the apex of ovulatory follicles prior to ovulation is a crucial step in the complex cascade of events initiated by the LH surge (Fata et al 2000, Ny et al 2002. Matrix metalloproteinases (MMPs) or matrixins, together with their tissue inhibitors (TIMPs) regulate ECM remodeling during both physiological and pathological conditions (Vu & Werb 2000, Coussens et al 2002, Curry & Osteen 2003.…”

“…However, very low levels of MMPs were present in the gland, whereas TIMP-1 was upregulated, shifting the proteolytic balance in favor of inhibition. The second proteinase-dependent stage begins around day 3 of involution and is characterized by a decrease in lactogenic hormones, collapse of glandular structures, and basement membrane destruction, resulting in active tissue remodeling and irreversible loss of the differenciated functions of the mammary gland (9,11,12,18). This phase is characterized by upregulation of mRNA and activity for proteolytic enzymes, including gelatinases A and B and stromelysin-1, and downregulation of the inhibitor TIMP-1 (18).…”
