Cell-free synthetic biology: Thinking outside the cell

Abstract: Cell-free synthetic biology is emerging as a powerful technology aimed to understand, harness, and expand the capabilities of natural biological systems without using intact cells. Cell-free systems bypass cell walls and remove genetic regulation to enable direct access to the inner workings of the cell. The unprecedented level of control and freedom of design, relative to in vivo systems, has inspired the rapid development of engineering foundations for cell-free systems in recent years. These efforts have le… Show more

“…The threat of a digital time bomb [138] also became an issue, in particular for "under-resourced institutions" (e.g., in the developing world). In general, printed paper is still seen as a robust long-term storage source, in particular for preserving the cultural heritage.…”

“…In vitro cell free expression systems that contain transcription and translation machinery provide the environment necessary for biologically-based technologies to function independently of living cells [122]. Originally utilized for in vitro protein expression, cell free expression systems have recently been recognized for their ability to support more complicated gene networks [123][124][125][126].…”

“…Further, the more precise control of conditions offered by in vitro expression systems has enabled the high-throughput tuning of synthetic constructs [123,124,127,128]. Despite these advancements, the application of cell-free methods has been limited to laboratory settings due to strict storage requirements, including the need for refrigeration.…”
