Cell culture monitoring via an auto‐sampler and an integrated multi‐functional off‐line analyzer

Abstract: Mammalian cell-based bioprocesses are used extensively for production of therapeutic proteins. Off-line monitoring of such cultivations via manual sampling is often labor-intensive and can introduce operator-dependent error into the process. An integrated multi-functional off-line analyzer, the BioProfile FLEX (NOVA Biomedical, Waltham MA) has been developed, which combines the functionality of three off-line analyzers (a cell counter, an osmometer, and a gas/electrolyte & nutrient/metabolite bio-profile analy… Show more

“…Moreover, adoption of PAT for continuous monitoring is encouraged by regulatory agencies as PAT should reduce process variability and provide a more consistent quality of product . Several efforts towards continuous monitoring of bioreactors have been made that utilize well known offline methods coupled with automated sampling . Derfus et al (2009) utilized an online sampler with a BioProfile FLEX system (NOVA Biomedical, Waltham MA) to continuously monitor nutrients/metabolites, cell concentration, and osmolality .…”

“…The industry standard instrument, the Bioprofile Analyzer, quantifies lactate concentration using an amperometric immobilized enzyme biosensor (Biomedical, 2016). Even though this analysis can be automated (Derfus et al, 2010), a large sample volume is required (at least 1 mL) and the lower limit of detection is in the millimolar range, limiting its application to later stages of culture. Enzymatic lactate detection kits, based on the lactate oxidase enzyme are also commercially available.…”

“…[4] Hence, controlling lactate and ammonia are common goals in cell culture process development. Glucose, lactate and ammonium are usually measured using commercially available multi-functional analyzers [5], and are usually measured using daily off-line samples during both development and commercial/clinical production batches.…”
