Celastrol, a Triterpene Extracted From Tripterygium wilfordii Hook F, Inhibits Platelet Activation

Abstract: Celastrol is an active ingredient of the traditional Chinese medicinal plant, Tripterygium wilfordii Hook F, which is known especially for its anti-inflammatory effects. However, on the cellular and molecular levels, celastrol's mechanism of action is only poorly understood. Because platelets contribute to inflammatory events, this study investigates the effects of celastrol on platelet function using flow cytometry, aggregometry, and adhesion assays. In in vitro experiments with human platelets, celastrol inh… Show more

“…Celastrol-A stock solution of celastrol (20 mg in 0.6 ml of dimethyl sulfoxide (DMSO; Sigma) was prepared and frozen at Ϫ20°C in small aliquots until needed, following the method described previously (31,32). The dose of celastrol (1 mg/kg) used in vivo in our study matched that used previously in different (non-arthritic) model systems (31,32).…”

“…Meanwhile, many studies have been dedicated to elucidating the potential molecular mechanisms underlying the anti-inflammatory and immunosuppressive effects of TEs [18], including the inhibition of platelet activation [20], the induction of nitric oxide [21], and prostaglandin E 2 [22] production. Based on studies both in vitro and in vivo , it is easy to speculate that TEs are likely to be types of herbal DMARDs, which differ from synthetic DMARDs.…”

“…Intraperitoneal injection of celastrol (1 to 3 mg/kg/day for 31 days), has anti-proliferative effects against prostate cancer in nude mice (Yang et al, 2006). Our recent study has demonstrated that celastrol inhibits platelet activation, adhesion to fibrinogen and aggregation in vitro, as well as fibrinogen binding of platelets in celastrol-treated mice, which may contribute to explain celastrol's anti-inflammatory and immunosuppressive effects (Hu et al, 2009). Furthermore, our previous data indicate that celastrol may also have anti-thrombotic properties.…”
