CD8+ T Cell-Based Molecular Classification With Heterogeneous Immunogenomic Landscapes and Clinical Significance of Clear Cell Renal Cell Carcinoma

Abstract: The tumor microenvironment (TME) exerts a high impact on tumor biology and immunotherapy. The heterogeneous phenotypes and the clinical significance of CD8+ T cells in TME have not been fully elucidated. Here, a comprehensive immunogenomic analysis based on multi-omics data was performed to investigate the clinical significance and tumor heterogeneity between CD8+ T cell-related molecular clusters. We identified two distinct molecular clusters of ccRCC (C1 and C2) in TCGA and validated in E-MTAB-1980 cohorts. … Show more

“…Using the NTP method, a total of eight cohorts obtained from distinct platforms were executed to measure and evaluate the prediction confidence for each patient, including GSE14520 ( Figure 5A ), ICGC-LIRI ( Figure 5C ), E-TABM-36, GSE76427, GSE25097, GSE104580, GSE116174, and GSE144269 ( Supplementary Figures S4A–F ). Consistent with a previous study, patients with false discovery rate <0.05 were detected for a subsequent analysis ( 36 ). In GSE14520 and ICGC-LIRI cohorts, Kaplan–Meier and multivariate Cox regression analyses were utilized to further elucidate the prognostic implications of the three clusters.…”

“…Tumor immune cell infltration has been approved to be associated with the prognosis of ccRCC and the response to immunotherapy [47,48]. Te importance of some infltrating immune cells has been confrmed, including regulatory T cells, CD8 T cells, NK cells, and resting mast cells [49][50][51][52]. Te immune cell infltration analysis between high and low NPEPL1 expression also revealed diferences in immune cells similar to previous studies.…”

“…Infiltrating immune cells in the tumor microenvironment have been uncovered to play prominent and respective roles in the biological behaviors of various cancers ( Mattei et al, 2012 ; Miyazaki et al, 2018 ; Georgouli et al, 2019 ). For example, high infiltration of CD8 + T cells usually inhibited cancer progression and indicated better prognosis in most cancers, but was associated with poor prognosis in clear cell renal cell carcinoma ( Qi et al, 2020 ; Wu et al, 2021 ). It has been revealed that identifying subtypes of the immune microenvironment in PaCa provides promising opportunities for therapeutic development based on the personalization of systemic immunotherapies ( Danilova et al, 2019 ).…”
