CD4+ and CD8+ T cells play a central role in a HDM driven model of allergic asthma

Abstract: BackgroundThe incidence of asthma is increasing at an alarming rate and while the current available therapies are effective in the majority of patients they fail to adequately control symptoms at the more severe end of the disease spectrum. In the search to understand disease pathogenesis and find effective therapies animal models are often employed. As exposure to house dust mite (HDM) has a causative link, it is thought of as the allergen of choice for modelling asthma. The objective was to develop a HDM driv… Show more

“…The main changes in the lungs include intense leukocyte and eosinophilic infiltration, airway hyperresponsiveness (AHR), mucus hypersecretion, and overexpression of T helper 2 (Th2) cell type cytokines including interleukin (IL)-4, IL-5, and IL-13 [5]. Th2 cell type cytokines are critical in allergic asthma by promoting the productions of immunoglobulin E (IgE) and various inflammatory mediators, as well as inducing eosinopoiesis [6]. Innate lymphoid cells (ILCs), derived from common lymphoid progenitor, are a group of innate immune cells in the lymphoid lineage [7].…”

“…Mice were terminally anaesthetised with urethane (10 μL·g −1 25% w / v i.p), bronchoalveolar lavage fluid (BALF) was collected as previously described (Raemdonck et al, ). Total and differential cell counts were performed as previously described (Raemdonck et al, ). Citrated blood was collected via cardiac puncture from mice terminally anesthetized with urethane (10 μL·g −1 25% w / v i.p.).…”

“…Although not dissimilar to the well-described heterogeneity of CD4+ T cells, the functional heterogeneity of CD8+ T cell subpopulations involved in different phases of the development of an allergic airway response has been less well defined. There is increasing evidence pointing to the contribution of CD8+ T cells to allergic airway responses in asthmatics and mice [5663]. Following sensitization and allergen exposure, mice deficient in the CD8α chain developed less airway inflammation and AHR compared to WT mice [59] and this was associated with decreased levels of the type 2 cytokine IL-13 in BAL fluid.…”
