CD14 C260T promoter polymorphism and the risk of cerebrovascular diseases: a meta-analysis

Abstract: Cerebrovascular diseases (CVD) are dysfunctions of the brain, resulting from diseases of blood vessels supplying the brain. Atherosclerosis is one of the major underlying causes of CVD, in which inflammation plays a crucial role. One of the inflammatory mechanisms contributing to atherogenesis is the activation of monocytes and macrophages, which could be mediated by the bacterial endotoxin lipopolysaccharide (LPS) via its receptor CD14. The C260T (rs2569190) single-nucleotide polymorphism (SNP) in the promote… Show more

“…The statistical heterogeneity was computed with the Q-statistic [46] (homogeneity rejected if P Q < 0.1 and/or I 2 > 75%) [47,48]. We calculated both Mantel-Haenszel (MH) (fixed effect (FE)) and random effects (RE) [49][50][51]. The effect was tested for the effect of the heterozygosity and for the dominant and the recessive models as well.…”

“…In 2009, Banerjee41 conducted a meta-analysis on the association between CD14 rs2569190 C>T polymorphism and cerebrovascular diseases but did not conduct a subgroup analysis on IS susceptibility. Thereafter, Misra et al conducted a meta-analysis with only six published case–control studies and failed to find any significant association between CD14 rs2569190 C>T polymorphism and IS susceptibility 42.…”

“…CD14 plays a major role in pathogen-activated signal transduction pathways and in the production of inflammatory cytokines [32]. CD14_IK- rs2569190 has been associated with prostate cancer in African Americans [33],and with coronary artery and cerebrovascular diseases [34], [35]. PRF1 codes for perforin 1, one of the main toxic proteins of cytolytic granules and a key effector in T-cell- and natural killer-cell-mediated cytolysis.…”
