CCL5-armed oncolytic virus augments CCR5-engineered NK cell infiltration and antitumor efficiency

Abstract: BackgroundNatural killer (NK) cells have potent antitumor activities. Nevertheless, adoptive transfer therapy of NK cells has gained very limited success in patients with solid tumors as most infused NK cells remain circulating in the peripheral blood instead of entering tumor sites. Chemokines and their receptors play important roles in NK cell distribution. Enhancing chemokine receptors on immune cells to match and be driven to tumor-specific chemokines may improve the therapeutic efficacy of NK cells.Method… Show more

“…On the other hand, imatinib upregulated some genes that could finally exert an “antiviral” action by sustaining the host immunological control of the viral attack, such as CD28 ( 35 ), Interferon gamma ( IFN gamma ) ( 41 ), C-C Motif Chemokine Ligand 5 ( CCL5 ) ( 33 ), C-C chemokine receptor type 5 ( CCR5 ) ( 34 ), and Toll-like receptor 3 ( TLR3 ) ( 43 ). Particularly interesting is CD28 , a co-stimulatory molecule also used for CAR-T production ( 55 ).…”

“…Inducing the expression of chemokine receptors on CAR-NK cells can dramatically improve tumor homing, as demonstrated for CXCR4 and CXCR1, co-expressed on EGFRvIII-specific or NKG2D ligand-targeting CAR-NK cells, respectively [156,157]. Infiltration of adoptively-transferred conventional NK cells into solid tumors is also augmented by induced expression of chemokine receptors [158,159].…”

“…A strong positive correlation between the expression of CCL5 and the infiltration of NK cells into human melanoma biopsies as well as various other solid tumors ( 47 ) was previously demonstrated. Survival of patients with melanoma with a high expression of CCL5 was increased ( 7 , 8 ).…”
