Caveolin‐1 deficiency induces a MEKERK1/2‐Snail‐1‐dependent epithelial–mesenchymal transition and fibrosis during peritoneal dialysis

Abstract: Peritoneal dialysis (PD) is a form of renal replacement therapy whose repeated use can alter dialytic function through induction of epithelial–mesenchymal transition (EMT) and fibrosis, eventually leading to PD discontinuation. The peritoneum from Cav1−/− mice showed increased EMT, thickness, and fibrosis. Exposure of Cav1−/− mice to PD fluids further increased peritoneal membrane thickness, altered permeability, and increased the number of FSP-1/cytokeratin-positive cells invading the sub-mesothelial stroma. … Show more

“…Numerous experimental models of peritoneal fibrosis have demonstrated that treatments directed against MMT, or against the MMT‐promoting stimuli, reduce the accumulation of myofibroblasts and ameliorate submesothelial fibrosis and angiogenesis, improving peritoneal structure and function . Thus, it is tempting to speculate that MMT may also be a therapeutic target to diminish the formation of PAs.…”

“…MCs were classified into epithelioid (E) or non-epithelioid (NE) phenotype according the Elliptical Factor (EF) which represents the relation between the major and the minor cell axis57. The dimensions of ten cells, randomly chosen in each culture, were measured using the ImageJ software (National Institutes of Health, Bethesda, Maryland, USA).…”

“…Therefore, disruption of miR-21 inhibited EMT at least by targeting SPRY2 and subsequently attenuating ERK1/2 pathway in ovarian cancer cells. Previous studies showed that ERK1/2 is a positive regulator of EMT in various human cancer 23, 24. TGFβ also induces EMT in a variety of human cancers.…”
