Caveolin‐1, cancer and therapy resistance

Abstract: Resistance of solid tumors to chemo- and radiotherapy remains a major obstacle in anti-cancer treatment. Herein, the membrane protein caveolin-1 (CAV1) came into focus as it is highly expressed in many tumors and high CAV1 levels were correlated with tumor progression, invasion and metastasis, and thus a worse clinical outcome. Increasing evidence further indicates that the heterogeneous tumor microenvironment, also known as the tumor stroma, contributes to therapy resistance resulting in poor clinical outcome… Show more

“…CAV1 codes for caveolin-1, a protein involved in promoting cell cycle progression [23]. UBE2I is a ubiquitin conjugating enzyme E2I, involved in protein degradation mechanism, while it can also stimulate transcription [24].…”

“…Src kinases mediate CAV1 phosphorylation at Tyrosine 14 (P-Y14) and of dynamin-2, both required for caveolae-mediated endocytosis and trafficking [13]. Depletion of CAV1 and the resultant reduction in the number of caveolae have been recently associated with a broad range of disease states such as cancer and cardiovascular and pulmonary diseases [14][15][16].…”

“…These results demonstrate that proper modulation of Cav-1 expression could be a supportive therapy in application of cytotoxic drugs. Modulation of Cav-1 as a supportive therapy in cancer treatment is clearly recognized and intensively discussed during the recent past [72,103,104]. At the same time, drug resistance in treatment of dermatological conditions was until now not directly connected with Cav-1 expression.…”
