This study showed that enzymatic interesterification of Amazonian oils could be an important tool in order to produce new oils with physicochemical properties that improve the applications of these raw materials. Structured oils of Amazonian patauá oil and palm stearin using two lipases were produced in three different enzymatic systems: first, a crude lipase from the fungus Rhizopus sp (a microorganism isolated in our laboratory); second, a commercial lipase; and third, to check any synergistic effect, a mixture of both lipases (Rhizopus sp and commercial). The lipase from Rhizopus sp was specific in the incorporation of oleic acid at the sn-1,3 positions of the triacylglycerol, resulting in an oil richer in saturated fatty acid in the sn-2 position. This enzyme, produced by solid-state fermentation, even though crude, was fatty acid and positional specific and able to operate at low concentration (2.5 %, w/w). In the second enzyme system, the commercial lipase from Thermomyces lanuginosus was not specific in the tested conditions; there was no change in the distribution of saturated and unsaturated fatty acids in the three positions of the triacylglycerol profile, there was only a replacement by the type of fatty acid at the same position. In the third enzyme system, the mixture of both lipases shows no synergic effect. The structured oils retained the concentration of bioactive α-and γ-tocopherol in the three enzyme systems.Triacylglycerol classes and Thermal behavior tests indicated the formation of more homogeneous triacylglycerols, especially the mono and di-unsaturated.

Vegetable oil-based fuels are promising alternative fuels for diesel and light fuel engines because of their environmental and economic strategic advantages. In this study, Ongokea gore oil (OGO) and its fully hydrogenated oil were transesterified by means of ethanol in the presence of sodium ethoxide. Fatty acid ethyl esters (FAEE) products were confirmed by 1 H NMR and characterized by physical-chemical methods in accordance with the ASTM D 6751 and AFNOR M 15-009 specifications for biodiesels and light biofuels. These methods concern determination of color, density, viscosity, flash and pour points, ash, water and sulfur contents, and corrosion on copper. It was found that pure fatty acid ethyl esters of Ongokea gore oil (B100) and its hydrogenated oil (B100-H) meet standard requirements for most of the biodiesel characteristics studied. Only the kinematic viscosity and density values were outside recommended biodiesel standard limits which makes them unsuitable for use in diesel engines. In accordance with the AFNOR M 15-009 specifications of light fuels, they can be used in light fuel engines. Physical-chemical properties of B20, a FAEE blend in petrodiesel, are within the limits prescribed for petrodiesel standards. In brief, Ongokea gore seeds, a nonedible and high-oil-producing feedstock, are suitable starting material for production of light biofuel. The latter blends in petrodiesel can be used as fuel in diesel engines.

Aos colegas e amigos dos demais laboratórios em especial: Tiago, Adriel, Rafael e Isac não só pelos encontros de diversão, mas pelos momentos de discussão em torno da pesquisa. Diversos biomateriais podem ser aplicados como suportes na imobilização de células totais de fungos filamentosos ou enzimas isoladas, visando a manutenção e o prolongamento da atividade enzimática em processos biocatalíticos. Exemplos promissores de biomateriais são a fibroína da seda e o alginato de sódio. A fibroína é um material protéico com alta estabilidade térmica, elasticidade, resistência à tensão, não sofre ataque microbiano, baixo custo de purificação e alta tenacidade, o alginato é um biopolímero versátil, devido a suas propriedades gelificantes em soluções aquosas. Assim, neste trabalho empregou-se micélios do fungo derivado de ambiente marinho, Penicillium citrinum CBMAI 1186, livres e imobilizados em biopolímeros (fibra de algodão, fibra de fibroína da seda e fibra de paina) na biorredução quimiosseletiva, regiosseletiva e enantiosseletiva da ligação α,β-C=C de enonas α,β-, α,β,γ,δ-e di-α,β-insaturadas previamente sintetizados pela a reação de condensação aldólica. Foi possível a utilização do fungo P. citrinum CBMAI 1186 na redução quimiosseletiva, regiosseletiva e enantiosseletiva da ligação dupla carbono-carbono de sistemas α,β-insaturados. A imobilização do fungo P. citrinum CBMAI 1186 em biopolímeros (algodão, fibroína da seda, paina e quitosana) permitiu a prolongamento da atividade celular do fungo. O protocolo desenvolvido foi capaz de obter compostos até então descritos apenas por síntese clássica. Também foi realizado reações de resolução enzimática de derivados de haloidrinas por diferentes lipases microbianas de: Pseudomonas fluorescens, Candida cylindracea, Rhizopus niveus e Aspergillus niger. A lipase de P. fluorescens foi imobilizada em esferas de fibroína do bicho da seda (método 1, via adsorção) e em blenda com alginato de cálcio (método 2, via encapsulação) em diferentes condições, tais como, variação de solvente, variação da quantidade de enzima imobilizada e tempo de reação. As condições otimizadas foram empregadas em diferentes haloidrinas, rendendo elevados excessos enantioméricos (ee > 99%) e alta razão enantiomérica (E > 200) para os produtos acetilados. Foi possível desenvolver um protocolo simples, barato e prático para a síntese enantiosseletiva de haloidrinas reforçando a versatilidade da fibroína e do alginato como suportes de imobilização para catalisadores heterogêneos. Também foi possível utilizar a lipase imobilizada (método 2) na reação de transesterificação para obtenção do biodiesel etílico. As melhores condições para o bom funcionamento do biocatalisador foram: 30% do biocatalisador, 20% de n-hexano, relação óleo e etanol de 1:4 a 32 ºC por 48 h em agitação magnética (400 rpm). Essas condições permitiram a formação de 42% de rendimento do biodiesel etílico.
