Caspase-6 mediates resistance against Burkholderia pseudomallei infection and influences the expression of detrimental cytokines

Abstract: Caspase-6 is a member of the executioner caspases and known to play a role in innate and adaptive immune processes. However, its role in infectious diseases has rarely been addressed yet. We here examined the impact of caspase-6 in an in vivo infection model using the Gram-negative rod Burkholderia pseudomallei, causing the infectious disease melioidosis that is endemic in tropical and subtropical areas around the world. Caspase-6-/- and C57BL/6 wild type mice were challenged with B. pseudomallei for comparing… Show more

“…This finding can support the positive effect in 5xFAD/C6-KO mice on memory task performance as plaque burden is associated with hippocampal memory [33,34]. IL-10 is an important negative immune regulator that can prevent damage caused by pro-inflammatory processes, and it was found to be in close relationship with caspase-6 [25]. In both caspase-6 KO groups, IL-10 gene expression was elevated compared to Het groups ( Figure 5C,D).…”

“…An additional cytokine of anti-inflammatory nature, IL-10, is an important negative immune regulator that can counteract damage caused by excessive inflammation [25,62]. In both caspase-6 KO groups, IL-10 was found to be elevated compared to non-KO groups.…”

“…The second gene, caspase-6 (LOC100652135) controls cell-death and the inflammation response (Cohen, 1997;Bartel et al, 2017). This gene is more highly methylated in head and brain tissue throughout development, compared to gametes (supplementary Fig.…”

“…Type III secretion is important in the interaction between B. pseudomallei and NLRC4 [44]. Recently, capsase-6 has also been shown to be important in response to B. pseudomallei, where capsase-6 deficient macrophages were shown to be less bactericidal and capsase-6 mice exhibited higher sensitivity alongside raise IL-1β and IL-10 production [45].…”
