“…Human A431, murine BALB/c‐3T3, and SVT2 fibroblasts were obtained from ATCC and were maintained in culture as previously described . For dose–response experiments, cells were seeded in 96‐well plates at a density of 2.5×10 3 cells per well; 24 h after seeding, increasing amounts of DiRu‐1‐encapsulated AFt (0.15–3 μ m ) or DiRu‐1 (0.05–0.2 μ m ) were added to the cells for 48 h. Cell viability was then assessed by MTT [3‐(4,5‐dimethylthiazol‐2‐yl)‐2,5‐diphenyltetrazolium bromide] assay, as described by Ferraro et al .…”

“…[13] Cell culture and cell growth inhibition Human A431, murine BALB/c-3T3, and SVT2 fibroblasts were obtained from ATCC and were maintained in culture as previously described. [62] For dose-response experiments, cells were seeded in 96-well plates at ad ensity of 2.5 10 3 cells per well;2 4h after seeding, increasing amounts of DiRu-1-encapsulated AFt (0.15-3 mm)o rD iRu-1 (0.05-0.2 mm)w ere added to the cells for 48 h. Cell viability was then assessed by MTT [3-(4,5-dimethylthiazol-2-yl)-2,5diphenyltetrazolium bromide] assay, [48] as described by Ferraro et al [6] Cell survival was expressed as the percentage of viable cells in the presence of test compound relative to controls. Twog roups of cells were used as controls:u ntreated cells and cells supplemented with identical volumes of buffer.E ach sample was tested in three independent analyses, each carried out in triplicate.…”

“…HaCaT cells were seeded at a density of 3.5 × 10 4 cells/cm 2 (100 mm eukaryotic cell plates) in complete medium for 24 h and then treated with 10 µg/mL of MeDHICA-melanin for 120 min. After UVA irradiation, cells were kept at 37 • C for 90 min, before performing the thiobarbituric acid reactive substances (TBARS) assay as described [48]. Briefly, cells were detached and 5 × 10 4 cells suspended in 0.67% TBA containing 20% trichloroacetic acid (TCA) (1:1 v/v).…”
