Carotenoid composition and changes in expression of carotenoid biosynthetic genes in tepals of Asiatic hybrid lily

Abstract: To elucidate the regulatory mechanisms of carotenoid accumulation in tepals of Asiatic hybrid lily (Lilium spp.), the content and composition of carotenoids and transcriptional changes of carotenoid biosynthetic genes were compared between tepals of yellow, red, pink and white flowers. Yellow and red tepal cultivars accumulated high amounts of carotenoids, and cultivars with pink and white tepals accumulated small amounts of carotenoids. Most of the carotenoids in yellow petals were antheraxanthin, (9Z)-violax… Show more

“…It should be noted that the accuracy of data may vary among manuscripts. antheraxanthin, violaxanthin, and lutein, while the major carotenoid in the red tepals of the cultivar 'Saija' is capsanthin 47 . The red ketocarotenoid capsanthin is found in the fruits of red pepper 2 but is rarely found in flower petals.…”

“…To amplify phytoene desaturase (PDS) fragments from N. benthamiana and white lupin, degenerate primers 5′-GCA AGR GAYGTTYTDGGWGG-3′ (forward) and 5′-RTTRCCA TCY AARAADGCC AT-3′ (reverse) were designed, respectively, from the amino acid sequences ARDVLGG and MAFLDGN conserved in plant PDS genes (Yamagishi et al 2010). In all, 505-bp fragments were amplified by reverse transcription (RT)-PCR, cloned into the pGEM T-Easy vector (Promega, Tokyo, Japan), and then sequenced.…”

“…Moreover, carotenoids are associated with flower colors, which may change with the relative proportions of individual carotenoids. For example, the main carotenoid in red lily flower petals is capsanthin, whereas a combination of violaxanthin, lutein, antheraxanthin, and β-carotene was observed in pink lily flowers (Yamagishi et al, 2010). The ratio of transcript abundance of specific carotenoid biosynthetic genes is probably important for the determination of flower color.…”
