Cannabinoid CB2 receptor‐mediated regulation of impulsive‐like behaviour in DBA/2 mice

Abstract: BACKGROUND AND PURPOSEThis study evaluated gene expression differences between two mouse strains, characterized by opposite impulsivity-like traits and the involvement of the cannabinoid CB2 receptor in the modulation of impulsivity. EXPERIMENTAL APPROACHBehavioural tests were conducted to compare motor activity, exploration and novelty seeking, attention and cognitive and motor impulsivity (delayed reinforcement task: session duration 30 min; timeout 30 s) between A/J and DBA/2 mice. Expression of genes for d… Show more

“…This is consistent with our previous findings that systemic or local administration of JWH133 into the nucleus accumbens inhibits cocaine self-administration (22), and that transgenic up-regulation of brain CB 2 Rs attenuates cocaine self-administration and cocaine-induced increased locomotion (19). The present findings are also congruent with recent reports that brain CB 2 Rs have important roles in other DArelated functions and/or CNS disorders (12,25,51,52).…”

“…More recent studies using RT-PCR also detected CB 2 mRNA in the cortex, striatum, hippocampus, amygdala, and brainstem (9)(10)(11)(12)(13)(14). Immunoblot and immunohistochemistry (IHC) assays detected CB 2 R immunoreactivity or immunostaining in various brain regions (13,(15)(16)(17)(18)(19)(20).…”

“…Coronal brain sections (500 mm) beginning at plates 19-20 (Paxinos and Franklin, 2001) were obtained in a cryostat ( À 10 1C). The VTA was microdissected according to a modification of the Palkovits method (Palkovits, 1983) as previously described (Navarrete et al, 2012). Total RNA was isolated from brain tissue micropunches using Trizol reagent (Invitrogen, Madrid, Spain) and subsequently retrotranscribed to cDNA.…”

“…In DBA/2J mice, which show high trait impulsivity, a CB2 agonist decreased impulsive responding and delay discounting, while a CB2 antagonist decreased novelty-seeking behavior and improved prepulse inhibition (PPI) [60]. In another study, the anticonvulsant and mood stabilizer topiramate reduced novelty-seeking behavior, impulsive responding and delay discounting in DBA/2J mice [61].…”
