Transient global cerebral ischemia (TGCI) induces by bilateral common carotid artery occlusion (BCCAO), and it mediates neuronal cell death of the CA1 hippocampal area. AM251 is a cannabinoid receptors type 1 (CB1) blocker that has been known to be protective against transient focal cerebral ischemia. JWH-015 is a selective agonist of CB2 and activator of CB1 that is involved in promotion of neuronal recovery and survival. The role of combined application of JWH-015 with AM251 in the rat model of GCI has been surveyed in this study. Male Wistar rats underwent 20 min of ischemia followed by reperfusion. Then, 1 mg/kg JWH-015 and 2 mg/kg AM251 were administered through caudal vein. The groups were control, sham, ischemia, vehicle, AM251, JWH-015 and AM251 + JWH-015. Animals were sacrificed at 14 days after reperfusion. The AM251 + JWH-015 group showed a significant increase in the protein expressions of AKT1, Bad 14-3-3, Bcl-2 and Bcl-XL, but it showed a considerable decrease in the protein expressions of Bad and JNK1/2 (p ≤ 0.05 vs. AM251, and JWH-015 groups). The AM251 + JWH-015 group had a significant higher number of alive cells and lower number of TUNEL-positive cells in the CA1 hippocampal area, and it also had a considerable improvement of spatial memory (p ≤ 0.05 vs. AM251, and JWH-015 groups). The results of this study showed that combined application of AM251 and JWH-015 could be neuroprotective against detrimental effects of ischemia probably via suppression of neuronal apoptosis and maintenance of their survival.

Prolonged urinary cannabinoid excretion in chronic frequent cannabis users confounds identification of recent cannabis intake that may be important in treatment, workplace, clinical, and forensic testing programs. In addition, differentiation of synthetic Δ9-tetrahydrocannabinol (THC) intake from cannabis plant products might be an important interpretive issue. THC, 11-hydroxy-THC (11-OH-THC) and 11-nor-9-carboxy-THC (THCCOOH) urine concentrations were evaluated during previous controlled cannabis administration studies following tandem alkaline/E. coli β-glucuronidase hydrolysis. We optimized recombinant β-glucuronidase enzymatic urinary hydrolysis before simultaneous liquid chromatography tandem mass spectrometry (LC-MS/MS) quantification of THC, 11-OH-THC, THCCOOH, cannabidiol (CBD), cannabinol (CBN), cannabigerol (CBG), tetrahydrocannabivarin (THCV) and 11-nor-9-carboxy-THCV (THCVCOOH) in urine. Enzyme amount, incubation time and temperature, buffer molarity and pH were optimized using pooled urine samples collected during a National Institute on Drug Abuse, Institutional Review Board-approved clinical study. Optimized cannabinoid hydrolysis with recombinant β-glucuronidase was achieved with 2000 IU enzyme, 2 M pH 6.8 sodium phosphate buffer, and 0.2 mL urine at 37°C for 16 h. The LC-MS/MS quantification method for hydrolyzed urinary cannabinoids was validated per the Scientific Working Group on Toxicology guidelines. Linear ranges were 1-250 μg/L for THC and CBG, 2-250 μg/L for 11-OH-THC, CBD, CBN, THCV and THCVCOOH, and 1-500 μg/L for THCCOOH. Inter-batch analytical bias was 92.4-112.4%, imprecision 4.4-9.3% CV (n = 25), extraction efficiency 44.3-97.1% and matrix effect -29.6 to 1.8% (n = 10). The method was utilized to analyze urine specimens collected during our controlled smoked, vaporized, and edible cannabis administration study to improve interpretation of urine cannabinoid test results.

A Neurociência, ciência responsável pelo estudo do sistema nervoso, têm crescido exponencialmente nas últimas décadas. Visto que muitos assuntos relacionados ao tema fazem parte do cotidiano popular, a divulgação científica das neurociências se torna relevante. Desta forma, o Grupo de Estudos em Neurociências da Universidade Estadual do Norte do Paraná (GEN / UENP) aderindo evento mundial de divulgação neurocientífica “Brain Awareness Week” (DANA Foundation), organizou a III Semana do Cérebro da UENP. O objetivo do presente trabalho é relatar sobre a ação presencial da III Semana do Cérebro da UENP, realizada em 2017, mostrando como um evento extensionista pode contribuir na formação dos alunos envolvidos e como ele integra ensino, pesquisa e extensão, tripé que é premissa da Universidade. Em dois dias de evento foram trabalhados diversos temas das neurociências que envolveram apresentação de filmes, palestras e discussões em forma de debate. Mais de 300 pessoas foram alcançadas com essa ação e pela quantidade de questionamentos levantados pelos participantes, podemos notar o interesse do público acerca da temática. A participação dos alunos do GEN como palestrantes aponta que eventos extensionistas podem ser utilizados como instrumento pedagógico importante na qualificação profissional dos acadêmicos.
