Cancer Stem Cells Accountability in Progression of Head and Neck Squamous Cell Carcinoma: The Most Recent Trends!

Abstract: Cancer stem cells (CSCs) play a major role in local recurrence and metastatic spread in head and neck squamous cell carcinomas (HNSCC). Evidence suggests that cancer stem cells are resistant to conventional therapy. So the emerging concepts of the role of cancer stem cells in the pathobiology of HNSCC should be understood carefully to be able to create new paradigms in treatment plans.

“…The number of tissue‐specific stem cells is lower when compared to other cells. This is consistent with the hypothesis that the CSC has their origin in the specific tissue stem cells . We have identified a distinct ALDH + and ALDH − population of HNSCC tumours.…”

“…In accordance with the American Association for Cancer Research (AACR), neoplastic stem cells or CSC are defined as cells from the tumour with capacity to self‐renewal as well as to differentiate into various types of cells of the same line, ensuring phenotypic heterogeneity of the tumour. According to the “cancer stem cell hypothesis,” malignant stem cells are located at the top of the tumour cell hierarchy, as they are the only ones that exhibit proliferative properties …”

“…There are several signaling pathways (Notch, Wnt/β-Catenin, and Sonic Hedgehog) and molecules (Oct-4, bone morphogenic protein) that regulate self-renewal ability of cancer stem cells and maintain stem cell proliferation (Pazarbaşı et al, 2011;Routray and Mohanty, 2014). The core stem cell signaling pathways Hh, Wnt, and Notch are deregulated in most cancers and act as survival pathways for CSCs (Ponti et al, 2006;Song et al, 2007).…”

“…EMT is a biological process enabling epithelial cells to acquire a migrant mesenchymal phenotype and is currently viewed as one of the essential steps during metastasis (Gupta and Massagué, 2006;Routray and Mohanty, 2014). In this process, epithelial cells lose cell-cell adhesion and cell polarity and decrease the expression of epithelial cells markers while upregulating mesenchymal markers such as vimentin, N-cadherin, and fibronectin.…”
