Cancer family history characterization in an unselected cohort of 121 patients with uveal melanoma

Abstract: Uveal melanoma (UM) is the most common primary intraocular malignancy in adults. The extent of the contribution of familial/hereditary predisposition to the development of uveal melanoma is largely unknown. Thus we sought to ascertain the frequency of cancers in patients with UM and their family members to identify the prevalence of hereditary/familial predisposition to cancer in these patients. An unselected series of 121 patients with UM seen in a university-based tertiary referral program were consented to … Show more

“…The recent paper [1], exploring the family history of cancer for persons diagnosed with uveal melanoma (UM) was aimed at elucidating a hypothesis that there may increased risk for some cancers in carriers of yet unidentified UM predisposition genetic alteration. While that is certainly a hypothesis worthy of further study, it has to be acknowledged that a familial predisposition can also be due to shared environmental factors.…”

“…Nearly all of the cancers among those diagnosed with UM and their first and second degree relatives listed in Table 3 of Abdel-Rahman [1] are vitamin D sensitive [6][7][8][9][10], possibly with the exception of prostate cancer in terms of incidence [11,12], although vitamin D status plays a role in survival for this cancer [13]. Still, diet and genetics certainly play important roles in risk for prostate cancer risk [12,14].…”

“…[1][2][3] Recent evidence suggests that patients with a cancer susceptibility may have higher frequencies of UM compared with the normal population. 4 Other risk factors are congenital ocular melanocytosis, melanocytoma, and neurofibromatosis.…”

“…Grant et al [1] commented on our recent manuscript on family cancer histories of unselected cohort of UM patients [2] and suggested that the familial predisposition observed in our study is likely due to shared environmental factors due to solar UVB dose and possibly Vitamin D status [1]. The authors cites ecological and observational studies suggesting such associations [3].…”
