Cancer chemopreventive activity of the prenylated coumarin, umbelliprenin, in vivo

Abstract: Umbelliprenin is a prenylated compound, which belongs to the class of sesquiterpene coumarins. In continuation of our earlier in-vitro finding, we determined to assess the cancer chemopreventive activity of umbelliprenin in vivo by using a two-stage carcinogenesis assay of mouse skin tumors induced by peroxynitrite as an initiator and TPA (12-O-tetradecanoylphorbol-13-acetate) as a promoter. In this assay, treatment with umbelliprenin along with peroxynitrite/TPA delayed the formation of papillomas up to week … Show more

“…Recently, there is a growing interest in the use of natural products for anticancer activity against cancer development (Ben Ammar et al 2009;KilaniJaziri et al 2009;Ben Sghaier et al 2011). These include fatty acids, vitamins, minerals, lichens, flavonoids and terpenoids (Chaudhary et al 2009;Iranshahi et al 2009;Ben Sghaier et al 2011;Ranković et al 2011). Terpenoids are one of the most important substances of this which are naturally occurring in plants that are thought to have positive effects on human and animal health (Hamułka et al 2012;Styrczewska et al 2012).…”

“…The plant compounds are widely used in treating cancer. Some of these compounds show anti tumor activity (Iranshahi et al, 2009;Sahebkar and Iranshahi, 2010). Bracken fern [Pteridium aquilinum (L.) kuhn (Dennstaedtiaceae)] as one of the most popular herbs on the planet (Taylor, 1990), is believed to carry such properties.…”

“…Despite the dominant cytotoxic activities of Umb against certain cancer cell lines (2,3,8), its proliferative activities have been observed in normal lymphocytes (2) and at lower doses in a colorectal cancer cell line in culture media (11), indicating that this compound may have different levels of cytotoxicity against different cancer cell lines in vitro.…”

“…This compound is able to suppress tumorigenesis through its anti-inflammatory, anti-genotoxicity, anti-invasive and lipoxygenase inhibitory activities (3)(4)(5)(6). In vitro, Umb has been reported to attenuate the proliferation of various cancer cell lines, and is able to prevent/delay cancer in in vivo animal models of papilloma and lung cancer (7)(8)(9). It hasbeen stated that the anti-tumor activities of Umb are partly due to a prenyl moiety (C7-OH) on the structure of the umbelliferone nucleus (3).…”
