Calcineurin Signaling and Membrane Lipid Homeostasis Regulates Iron Mediated MultiDrug Resistance Mechanisms in Candida albicans

Abstract: We previously demonstrated that iron deprivation enhances drug susceptibility of Candida albicans by increasing membrane fluidity which correlated with the lower expression of ERG11 transcript and ergosterol levels. The iron restriction dependent membrane perturbations led to an increase in passive diffusion and drug susceptibility. The mechanisms underlying iron homeostasis and multidrug resistance (MDR), however, are not yet resolved. To evaluate the potential mechanisms, we used whole genome transcriptome a… Show more

“…Genes associated with ergosterol biosynthetic pathway are regulated by iron availability in yeast (82). In agreement with the deregulated cellular iron homeostasis in the mutant, five genes in the ergosterol biosynthesis pathway were downregulated, with a commensurate decrease in ergosterol levels ( Fig.…”

“…That Candida cells are capable of displaying a metabolic shift has already been documented (1). For example, we observed previously that iron deprivation compels Candida cells to adapt to a fermentative metabolism (21). Recently, upregulation of glycolytic genes was reported as implicated in biofilm formation in a hypoxic environment; this process was regulated by Tye7p, a transcriptional activator of glycolytic genes helping cells to adapt to hypoxia (3).…”

“…Ten milligrams of purified total RNA was used to synthesize cDNA by using the protocol described previously (www.transcriptome.ens.fr /sgdb/protocols/labeling.yeast.php). We used a previously described direct-labeling method to label cDNA with Cy3 and Cy5 dyes (15). The labeled cDNA was mixed with hybridization buffer and applied immediately to microarray slides.…”
