c‐erbB‐2 and c‐erbA‐1 (ear‐1) Gene Amplification and c‐erbB‐2 Protein Expression in Japanese Breast Cancers: Their Relationship to the Histology and Other Disease Parameters

Abstract: We studied c-erbB-2 and c-erbA-1 (ear-1) gene amplification, and c-erbB-2 protein expression in 123 primary Japanese breast cancers. c-erbB-2 amplification was found in 19 of the 123 tumors (15%), and c-erbA-1 was coamplified in 7 of the 19. The presence or absence of c-erbB-2 amplification correlated with the grade of cellular atypism (P = 0.008), or that of mitotic index (P = 0.002), but not with the histologic types. The tumor size (P = 0.04) and the lymph node status (P = 0.06) were associated, but the pat… Show more

“…Slamon et al [16], Pauley et al [23] and Yamada et al [24] have reported that c-erbB-2 amplification is proportional to the number of lymph node metastases. There was a correlation between the degree of c-erbB-2 amplification and the stage of disease [25] in our data, but other reports have seen no correlation [17,24,26].…”

“…Although a statistical difference in the copy number of c-erbB-2 was not recognized in the groups with positive and negative distant metastases, or in the groups with positive and negative invasion to the pectoralis major muscle or skin, a tendency towards amplification of the c-erbB-2 gene was seen in patients with distant metastases and invasion to pectoralis major muscle or skin. Some reports have recognized a correlation between the degree of c-erbB-2 amplification and tumor size [26].…”

“…In the study of Rossi et al, the percentage of cases with HER-2/ neu protein overexpression (3+) and FISH negativity rose to 22% (38). In the literature it is proposed that these 3+ cases could be due to single copy overexpression of the HER-2/neu gene (39). These cases are considered clinically similar to the immunohistochemically HER-2/neu negative patients (40 (42)(43)(44).…”

“…cerbB-2 expression has also been correlated with nuclear alterations in several studies of human breast cancer (14,(17)(18)(19), showing the existence of a correlation between a morphological and a biological factor.…”
