Brimonidine reduces TGF-beta-induced extracellular matrix synthesis in human Tenon’s fibroblasts

Abstract: BackgroundBrimonidine is a highly selective α2 adrenergic agonist that has been widely used in anti-glaucoma eyedrops. The aim of this study was to investigate its putative anti-fibrotic role in the fibrosis caused by activated Tenon’s fibroblasts.MethodsPrimary cultured human Tenon’s fibroblasts were exposed to 2.0 ng/mL of transforming growth factor-β1 (TGF-β1) for up to 48 h. In the presence of various concentrations of brimonidine (from 0.0 to 10.0 μM), the expression levels of fibronectin, collagen types … Show more

“…Furthermore, we also identified that BRI induced significant inhibitory effects on the TGF-β2-related increases in the barrier function of 2D cells and the hardness of 3D spheroids. Similar to the BRI-induced inhibition of TGF-β2-related fibrosis in human Tenon’s fibroblasts in vitro [ 26 ], our results indicate that BRI causes TGF-β-stimulated ECM synthesis.…”

“…Myofibroblast formation is associated with fibrotic scars in various injury models 34 . TGF-β1 promotes IL-6 expression and induces trans-differentiation of human Tenon’s fibroblasts to myoblasts 35 . Therefore, we hypothesized that TGF-β1 also promotes IL-6 production in HCFs, inducing differentiation from cardiac fibroblasts to myofibroblasts.…”

“…Likewise, MSC transplanted into MPS VII mice cornea also acquired the characteristics of keratocytes. The transplanted cells supplied functional β-glucuronidase to degrade accumulated glycosaminoglycans thereby assisting in lysosome recycling ( 60 ). On the other hand, MSCs also secrete paracrine factors that exert anti-inflammation and anti-fibrosis functions, thus suppressing inflammatory neovascularization ( 61 , 62 ).…”
