“…THE AMOUNTS OF PROSTAGLANDINS extracted from various tumours, notably from human carcinomas of the breast (Bennett et al, 1975Powles et al, 1976), large intestine (Bennett et al, 1977) and kidney (Atkins et al, 1977) and from certain experimental neoplasms (Powles et al, 1 973;Tashjian et al, 1974;Galasko, 1.976;Galasko & Bennett, 1976) are usually greater than from the corresponding normal tissues. Prostaglandins and other lessclearly defined "tumour-associated" products may be involved in the establishment and growth of metastases in bone and possibly in other sites (Carter, 1978;Bennett, 1979).…”

“…We have previously reported (Powles et al, 1976) a higher incidence of bone metastases in patients whose tumour showed high in vitro osteolytic activity by hot concomitant culture method. The longer follow-up in this study shows that the apparent difference in the relapse and survival rates seen in the first 20 months was not maintained.…”

“…This destruction is probably mediated by release of osteolytic substances from tumour cells. Using in vitro organ culture (Reynolds, 1968) we previously reported that 23/38 primary human breast carcinomas showed significant osteolytic activity (Powles et al, 1976) mediated, at least in part, by prostaglandins (Dowsett et al, 1976;Powles et al, 1973;Goodson et al, 1974). Early clinical follow-up appeared to indicate that patients whose tumours produced the greatest in vitro osteolytic activity had the greatest risk of bone metastases (Powles et al, 1976).…”

“…Human breast cancers have been shown to produce prostaglandins PGE2 and PGF2a in vitro (Greaves et al, 1980;Rolland et al, 1980a;Bennett et al, 1980a;Dowsett et al, 1976) and raised levels are found in blood draining the tumours in patients (Stamford et al, 1980). Breast cancer explants in vitro produce osteolysis, partly by a PG-mediated mechanism (Powles et al, 1976;Bennett et al, 1975;Dowsett et al, 1976).…”
