“…These studies showed that S-100 protein was synthesized predominantly on free polysomes, but did not exclude an absolute requirement of a brainspecific factor for translation of s-100 protein messenger RNA. Although s-100 protein could be synthesized efficiently in a crossed reconstituted system of brain polysomes and a liver enzyme fraction [28] the results of these experiments were inconclusive since initiation and elongation factors are known to be associated with polysomes.…”

“…A postmicrosomal supernatant, prepared in 10 mM Tris-HC1, pH 7.6, 5.2 mM MgCI2, 0.2 mM EDTA, 1 mM DTT, and 0.3 M sucrose (Falvey and Staehelin, 1970) was diluted with 2 volumes of 1 mM DTT. The pH was then adjusted to 4.7 with 1 M acetic acid (Zomzely-Neurath and Roberts, 1972). After being stirred for 30 rnin at 4"C, the precipitate was collected by centrifugation at 34,500 X g for 15 min.…”

“…Soluble and ribosomal proteins were extracted from five pooled young and five pooled old mouse brains closely following the technique of Zomzely-Neurath and Roberts (42). 1 g of brain was homogenized in 9 ml of 50 niM Trizma-HCl (pH 7.4), 25 triAf KC1, 4 mM MgCI., 250 mM sucrose (TKMS), then centrifuged at 15,000 # for 15 min at 4 °C.…”
