Brain‐derived neurotrophic factor alleviates the oxidative stress induced by oxygen and glucose deprivation in an ex vivo brain slice model

Abstract: Brain‐derived neurotrophic factor (BDNF) is considered as a putative therapeutic agent against stroke. Since BDNF role on oxidative stress is uncertain, we have studied this role in a rat brain slice ischemia model, which allows BDNF reaching the neural parenchyma. Hippocampal and cerebral cortex slices were subjected to oxygen and glucose deprivation (OGD) and then returned to normoxic conditions (reperfusion‐like, RL). OGD/RL increased a number of parameters mirroring oxidative stress in the hippocampus that… Show more

“…The list of factors includes, among others, BDNF, HGF, IL-10, IL-1RA, IL-4 and LIF. Interestingly, all of them have been shown to have a protective role towards IRI damage of different organs, including the liver, brain, heart and kidney [ 52 , 53 , 54 , 55 , 56 , 57 ]. These findings point to the possibility of dissecting the precise molecular mechanisms that the different bioactive factors might activate to exert their function in liver IRI.…”

“…Interestingly, our analyses also indicate that genes involved in stress response are regulated differently in the substantia nigra compared with the rest of the nigro-striatal network. Different populations of neurons show various degrees of vulnerability to oxidative stress (González-Rodríguez et al, 2019) and cerebellar granule cells and substantia nigra dopaminergic neurons have been reported…”

“…For instance, eye–brain electrochemical transduction and retinal nerve fiber alterations, structural and functional abnormalities in the endothelial progenitor cells, interlinks between mitochondrial energy production, reactive oxygen species availability (e.g., nitric oxide—NO), and ion homeostasis (e.g., calcium influx) seem to play initiating roles in a brain with migraine (Ashina, 2020 ; DeBuc et al., 2020 ; Lee et al., 2008 ; Polak et al., 2003 ; Zhang et al., 2001 ). Therefore, regulation and availability of signaling biomolecules (e.g., NO, and brain‐derived neurotrophic factor—BDNF) and regulation of signaling pathways (e.g., cAMP‐response element‐binding protein—CREB) are essential during health and illness, including the symptoms of migraine attacks, as stressful insults to the brain (González‐Rodríguez et al., 2019 ; Khosravi et al., 2019 ).…”
