Bovine Serum Albumin as a Lyoprotectant for Preparation of DNA Dry Powder Formulations Using the Spray-Freeze Drying Method

Abstract: The development of efficient and selective therapeutic gene delivery methods is a potential medical treatment for intractable diseases. Dry powder inhalers (DPIs) can efficiently deliver drugs locally to the lung. Many reports discuss preparation methods for DPIs. Spray-freeze drying is a method by which highly porous particulates can be prepared. However, altered physical properties after preparation may result in changes in gene expression. In this study, bovine serum albumin (BSA) was added as a lyoprotecta… Show more

“…Similar to the case of CHOL-containing formulations, this reflects a lowered electrostatic repulsion between the BSA-coated nanocarriers. A similar effect of BSA on ζ-potential has been observed for other cationic liposomes containing DOTAP, 86 although a different behavior was found upon full plasma incubation. 6 However, DOTAP-induced changes in BSA secondary structure have been reported, 32 and this effect was recently demonstrated to also occur in liposomes containing positively charged moieties, other than lipids, on their surface.…”

“…23) After preincubation for 10 min, the powder was sprayed on the apical side using a plastic syringe connected to a disposable micropipette tip. 24) At predetermined time intervals, 0.2 mL of TM was collected from the basal compartment for a period of 6 h, and was replaced with the fresh medium. Because the temperature in the nasal cavity is around 25-37°C, the temperature of the monolayer was maintained at 25-37°C throughout the experiment.…”

“…It is an advanced particle formation method in which a liquid material is intended to spray in hot air or nitrogen (act as a freezing medium) followed by lyophilization. The drying of thermolabile substances such as insulin, proteins, DNA, etc., can be obtained with this technique, supported by the preparation of delicate, porous particulate matter . The high degree of control over the residual moisture content, mass density, nano ranged particle size, easy manipulation of various parameters (i.e., temperature of the cryogenic fluid, concentration of the solution, atomizer type, speed, etc.)…”
