Body Adiposity and Endocrine Profile of Female Wistar Rats of Distinct Ages that were Early Weaned

Abstract: Early weaning (EW) is a risk factor for metabolic syndrome. Male rats that were precociously weaned present neonatal malnutrition and, in adulthood, developed overweight, accumulation of body fat, dyslipidemia, changes in glycemic homeostasis, hyperleptinemia, and increase of vitamin D. As metabolic profile of early-weaned females is not known, we investigated the endocrine-metabolic parameters in adolescence an… Show more

“…Recently, our group demonstrated that PEW females have reduced serum oestrogen levels in adulthood (Pietrobon & Bertasso et al . ), which could contribute to the reduction in insulin secretion in this group, despite the lack of alterations in ERα expression in islets. Epidemiological and experimental studies suggest that oestradiol protects insulin production in diabetic females (Louet et al .…”

“…Islets were isolated from fasted offspring at PN45 and PN180 by collagenase (0.8 mg/mL; Sigma‐Aldrich, St Louis, MO, USA, 5% bovine serum albumin (BSA) and 0.6% HEPES according to a previous report (Pietrobon et al . ). After digestion of the exocrine pancreas, the islets were collected with a micropipette under a microscope to exclude any contaminating tissues.…”

“…Female offspring in both pharmacological and NPEW models exhibited higher adiposity and hyperphagia, despite having normal body weight. A sexdependent mechanism seems to be involved in this phenotype because the females showed differences in hormonal profiles, 115 BAT thermogenic capacity, 88 fat deposit distribution, and glucocorticoid status. 111…”

“…Hyperphagia, higher body mass, and TG/leptin serum levels as well as leptin resistance outcomes were also observed in previous studies encompassing other animal models of early weaning. 10,11,29,41,42 Early-weaned rats presented higher LVM, IVS thickness and LVPW thickness, both in diastole, compared to respective adult and elderly controls. RWT was higher in this group at postnatal day 90 but not 150 or 365.…”
