Blood Plasma of Patients with Parkinson’s Disease Increases Alpha-Synuclein Aggregation and Neurotoxicity

Abstract: A pathological hallmark of Parkinson's disease (PD) is formation of Lewy bodies in neurons of the brain. This has been attributed to the spread of α-synuclein (α-syn) aggregates, which involves release of α-syn from a neuron and its reuptake by a neighboring neuron. We found that treatment with plasma from PD patients induced more α-syn phosphorylation and oligomerization than plasma from normal subjects (NS). Compared with NS plasma, PD plasma added to primary neuron cultures caused more cell death in the pre… Show more

“…e ELISA assay used for measuring pS-α-syn-RBC was established previously [25] and has been applied to detect pS-α-syn in aging monkey brains [24][25][26] and pS-α-syn formed in PD plasma [27]. e detection antibody was 3D5 mouse monoclonal anti-α-syn, which recognizes a sequence of 115-121 amino acids specific to human α-syn [23] and has been demonstrated previously for its specificity in detecting α-syn in human RBCs [16].…”

“…The ELISA assay used for measuring pS-α-syn-RBC levels was established previously and has been applied to detect pS-α-syn in monkey brains (Liu et al, 2015 ) and pS-α-syn formed in PD plasma (Wang et al, 2016 ). The detection antibody was 3D5 mouse monoclonal anti-α-syn, which recognizes a sequence of 115–121 amino acids specific to human α-syn (Yu et al, 2007 ) and has been well-characterized previously for its specificity in detecting α-syn in human RBCs (Wang et al, 2015 ).…”

“…Porcine choroid plexus produces several neurotrophins and can be safely delivered to the striatum in an encapsulated formulation to protect them from immune attack [ 63 ]. NTCELL is an alginate-coated capsule containing clusters of neonatal porcine choroid plexus cells [ 64 ]. In 2013, Living Cell technologies hosted a phase I/II trial of NTCELL implantation ( Table 2 ).…”
