Biotechnological production of centellosides in cell cultures of Centella asiatica (L) Urban

Abstract: Centella asiatica (L.) Urban plants have been used since ancient times for their medicinal properties, and their extracts have proven antioxidant, wound healing, sedative, and neuroprotective activities, among others. The natural compounds responsible for C. asiatica bioactivity are triterpene saponins formed from the dammarene branch of the triterpene biosynthetic pathway, collectively known as centellosides, with madecassoside and asiaticoside and their aglycones, madecassic acid and asiatic acid being the m… Show more

“…Centella asiatica cell line UBCA17 is characterized by a high capacity for biomass and centelloside production under elicitation [15]. The positive effects of treatments with elicitors such as MeJA, SA, copper ions, and YE on centelloside production have been previously demonstrated in both C. asiatica plant cell and hairy root cultures [7]. In this work, we corroborated the positive effect of MeJA, SA, and YE on centelloside biosynthesis, and also for the first time demonstrated the effectiveness of the new elicitor CORO in increasing centelloside production (more than fivefold), even at concentrations 100-fold lower than MeJA, without affecting the growth capacity of the cell cultures ( Fig.…”

“…Therefore, biotechnological production methods constitute an alternate and efficient way for obtaining specific secondary metabolites. The abatement in the diversity of plant by uncommon cutting and exploitation lead to various tissue culture methods to preserve the plant diversity and production of concerned products from it [33] .…”

“…PCCT is a powerful technology platform for production of bioactive phytochemicals, active cosmetic additives, food supplements and nutraceuticals, which in general utilizes non-genetically modified plant cells, grown under controlled microenvironment in a form of suspended single cells and/or small aggregates. Several pharmaceutically active compounds, including: tropane alkaloids atropine, hyoscyamine and scopolamine [4]; benzylisoquinoline alkaloid protopine [5]; diterpenoids as paclitaxel [6]; triterpenoids as ursolic and oieanolic acids [7,8]; pentacyclic triterpenoid saponins as centellosides [9]; stilbenoids as resveratrol [10]; aryl tretralin lactone podophyllotoxin [11]; phenylethanoid glycosides as verbascoside and leucosceptoside A [12]; etc. have been found to accumulate in plant cell suspensions.…”
