Bioluminescent Enzymatic Assay as a Tool for Studying Antioxidant Activity and Toxicity of Bioactive Compounds

Abstract: A bioluminescent assay based on a system of coupled enzymatic reactions catalyzed by bacterial luciferase and NADH: FMN-oxidoreductase was developed to monitor toxicity and antioxidant activity of bioactive compounds. The assay enables studying toxic effects at the level of biomolecules and physicochemical processes, as well as determining the toxicity of general and oxidative types. Toxic and detoxifying effects of bioactive compounds were studied. Fullerenols, perspective pharmaceutical agents, nanosized par… Show more

“…Changes in general toxicity and oxidative toxicity of aqueous media under exposure to humic substances were studied previously in [47][48][49]. Later [41,[50][51][52], the toxicity and antioxidant activity of a series of fullerenols (i.e., carbon nanostructures, water-soluble derivatives of fullerenes, perspective pharmaceutical agents) were evaluated and compared. Effects of MNPs and MNPs with modified surface on bacterial enzyme reactions have not been studied yet.…”

“…Decrease of both general and oxidative toxicities under addition of humic substances (products of natural decomposition of organic matter in soil and bottom sediments) was studied in [37] , [38] , [39] . Description of the enzyme bioluminescent technique to evaluate antioxidant activity and toxicity of bioactive compounds is presented in [40] . It should be noted that, similar to reactive oxygen species, active chlorine species can also produce toxic effects [41] .…”

“…Figure 11. Antioxidant activity of humic substances was studied in [80][81][82][83][84] and fullerenols -in [84][85][86][87]. Cellular and enzymatic assays were used to evaluate a General Toxicity (GT) under conditions of the oxidative stress; this stress was modeled in the solutions of exogenous organic or inorganic oxidizers (1,4-benzoquinone or potassium ferricianide, respectively).…”
