Biological Roles of the Two Components of Crotoxin

Abstract: Crotoxin, the Brazilian rattlesnake neurotoxin, generally behaves as a homogeneous protein; however, it is a molecular complex of an acidic and a basic protein. These can be separated after alkylation or acylation of the amino groups, or on carboxymethyl cellulose at pH 4, or on DEAE-cellulose in 6 M urea. The two proteins differ greatly in composition, but one or both may exist in the form of closely related variants. Their molecular weights appear to be about 8400 and 13,000.The acidic protein lacks the hemo… Show more

“…It is a complex of two distinct protein subunits [2,3] : a strongly basic component (component B, M , 12000) which carries the phospholipase activity and an acidic protein (component A, M , 10000) which has no enzymatic activity and is not toxic by itself. Whereas the isolated subunits are either not or only a little toxic, they recombine into a complex which exhibits the same toxicity as the original material [4,5].…”

“…En este trabajo se aisló e identificó la crotoxina B del veneno de C. d. cumanensis. Esta fosfolipasa A2 hace parte del complejo crotoxina, formado por la crotoxina A (subunidad ácida) y la crotoxina B (subunidad básica) unidas de forma covalente (29), y es responsable de la neurotoxicidad del veneno de las cascabeles suramericanas (30,31).…”

“…A well-characterized example of neurotoxic sPLA 2 myotoxin is crotoxin (CTX), a venom component of the South American rattlesnake Crotalus durissus terrificus (Hendon and Fraenkel-Conrat, 1971;Mebs and Ownby, 1990;Salvini et al, 2001). Among the non-neurotoxic sPLA 2 myotoxins, two different types are currently described: the "classical" Asp-49 PLA 2 s, which catalyze the hydrolysis of the ester Comparative Biochemistry and Physiology, Part C 143 (2006) 284 -294 www.elsevier.com/locate/cbpc bond in the sn-2 position of glycerophospholipids in a Ca 2+ dependent manner; and the "variant" Lys-49 PLA 2 s which have been described as devoid of enzymatic activity due to replacement of Asp-49 by Lys, together with other modifications in the calcium-binding loop (Lomonte et al, 2003b).…”
