Cytokines are thought to regulate prostaglandin (PG) secretion in the bovine endometrium. However, there is no consensus about the role of interleukin-1a (IL1A) on PG secretion. The objective of this study was to examine the influence of IL1A on basal and interferon-t (IFNT)-regulated PG in vitro secretion, as well its effects on PG secretion, progesterone (P 4 ) output, and corpus luteum (CL) in vivo lifespan. Explants of bovine endometrium (days 16-17 of the estrous cycle or early pregnancy) were stimulated with IL1A (10 ng/ml), IFNT (30 ng/ml), or IL1A combined with IFN. IL1A alone strongly stimulated luteotrophic PGE 2 secretion by endometrial tissues of both pregnant and nonpregnant cows. IL1A also stimulated luteolytic PGF 2a output in the late luteal phase. IFNT augmented the stimulatory effect of IL1A on PGE 2 secretion. In an in vivo experiment, saline or IL1A at different doses (0.001-10 mg/per animal) was applied to the uterine lumen on day 16 of the cycle. Only the highest dose of IL1A caused a temporal increase in PGF 2a secretion, while it had no effect on P 4 secretion or CL lifespan. Application of 0.1 and 1 mg IL1A stimulated P 4 and PGE 2 output and prolonged the CL lifespan. Although IL1A may stimulate in vitro luteolytic PGF 2a secretion during the estrous cycle, it only acts as a luteotrophic factor in vivo. IL1A increased luteotrophic PGE 2 and P 4 output, inhibiting spontaneous luteolysis. These luteotrophic effects may result in appropriate luteal development and function in cows during the estrous cycle and early pregnancy.

ABSTRACT. We developed a procedure for the large-scale purification of bovine interferon-τ (boIFN-τ) by means of a silkworm-baculovirus gene expression system. Recombinant boIFN-τ (rboIFN-τ) was efficiently produced in the silkworm infected with boIFN-τ cDNA recombinant baculovirus and accumulated in the haemolymph. To establish a purification method suitable for mass production, we tried three crude purification methods, namely, an acidification and neutralization treatment (ANT), silica gel column chromatography (SGCC), and Blue sepharose column chromatography (BSCC) with a combination of Q-sepharose (QSC) and chelating sepharose column chromatographies (CSCC). As a result, the acidification and neutralization treatment was found to be the most efficient and cost effective. With this combination, we obtained 91% pure products. To confirm the applicability of the procedure for mass production, we inoculated 100 silkworms with the recombinant virus, and recovered about 4.55 mg (1.26 × 10 8 U/mg) of 91% pure rboIFN-τ by means of a combination of the ANT, followed by QSC and CSCC. Sci. 66(11): 1395-1401, 2004 Interferon (IFN)-τ, originally named trophoblast protein-I, is the major secretory protein of trophoblastic tissues and plays an important role in the maternal-fetal recognition of pregnancy in ruminants [1,8,19]. Interferon-τ has been classified as a member of the large multi-gene type I IFN family, and has about 85 and 70% overall homology to IFN-ω on the bases in the cDNA nucleotide and amino acid sequences, respectively [4,9,20]. As expected from the homology, IFN-τ possesses most of the major functions of IFNs, including anti-viral activity, but does not lead to a febriferous reaction, a serious side effect of the IFN therapy. Because of its biological activities, IFN-τ is applied as a useful tool in ongoing studies on the establishment of pregnancy, and is also expected to be used as an agent to improve the pregnancy rate through the control of implantation and to treat viral diseases. For these applications, it is essential to establish a mass production procedure for purified IFN-τ. Up to now, recombinant ovine and bovine IFN-τ boIFN-τ) had been expressed by baculovirus, yeast and Escherichia coli (E.coli) gene expression systems [3,6,16]. Among these, the baculovirus gene expression system is the most suitable system for producing recombinant IFN-τ for animal experiments and practical biological agents because of its similarity in terms of post-translational modifications, such as glycosylation, phosphorylation, conformation, antigenicity and so on [14]. Bovine interferon (bIFN) τ plays a crucial role in maternal-fetal recognition and was expressed using a Bombyx mori (Bm) nuclear polyhedrosis virus (silkworm baculovirus) gene expression system. The biological effects of Bm-recombinant bIFNτ (rbIFNτ) on prostaglandin (PG) F2α synthesis were investigated in cultured bovine endometrial epithelial cells with oxytocin (OT, 100 nM) and on the in vitro development of bovine embryos. Bm-rbIFNτ and OT were shown to suppress PGF2α production in a dose-dependent manner. When in vitro produced morula stage embryos were cultured for 72 hr in modified CR1aa medium supplemented with or without rbIFNτ, Bm-rbIFNτ (10 ng/ml) significantly promoted development to the expanded blastocyst stage. In conclusion, Bm-rbIFNτ was suggested to have the same bioactivity as native IFNτ.
