Biological Activity of Recombinant Bovine Interferon .TAU. Using an Autographa california Nuclear Polyhedrosis Virus Expression System

Abstract: Abstract. Bovine interferon (bIFN) τ, which plays a key role in maternal-fetal recognition of pregnancy, was expressed by an Autographa californica nuclear polyhedrosis virus expression system. cDNA coding bIFNτ was derived from cultured trophoblast cells. The recombinant (r) bIFNτ had high antiviral activity (1 × 10 8 IU/mg) and the molecular weight of rbIFNτ was estimated to be 23 kDa by Western blotting analysis. We investigated the biological effect of rbIFNτ on prostaglandin (PG) F2α synthesis in cultured… Show more

“…The purpose of the present work was not to determine whether IFNT regulates basal PG production but only to use the lowest effective IFNT dose that can modulate IL1A-stimulated PG production exclusively. Therefore, according to Takahashi et al (2003) and Okuda et al (2004), we used IFNT at a relatively small dose of 30 ng/ml that shows bioactivity and was able to Figure 4 Concentrations of progesterone in peripheral blood plasma of cows infused with saline (gray bars) and various doses of IL1A (lines). All reagents were infused into the uterus.…”

“…Moreover, some differences in IFNT action on basal PG production depend on the type and biological activity of IFNT used in the studies as well as on diverse doses (Asselin et al 1997, Parent et al 2002, Takahashi et al 2003, Spencer et al 2007a, 2007b.…”

“…Bovine IFN-τ cDNA was prepared as reported by Takahashi et al [24]. The cDNA was excised and inserted into the baculovirus transfer vector (pBm050) to obtain the recombinant transfer vector (pBm boIFN-τ).…”

“…IFNτ binds to receptors (type I IFN receptor; IFNRI) on the uterine endometrium and suppresses transcription of the estrogen and oxytocin receptors genes to block pulsatile release of prostaglandin (PG) F 2α . Furthermore, it has been demonstrated that IFNτ inhibits PGF 2α synthesis by cultured endometrial epithelial cells [7, 11, 33, 38]. This allows for maintenance of corpus luteum function and the continuous production of progesterone [2, 3, 20].…”
