Biological activities of some Fluoroquinolones-metal complexes

Abstract: Background: Metal ions play a vital role in the design of more biologically active drugs. Aim: The paper reviewed the antimicrobial, toxicological and DNA cleavage studies of some synthesized metal complexes of fluoroquinolone antibiotics. and Methods: Literature searches were done using scientific databases. Results: Computer search was used to reveal relevant studies. Spectrophotometric and X-ray analyses of the metal complexes have revealed the bi-dentate coordination of fluoroquinolone ligand to the metal … Show more

“…As a fluorine atom at position 6 of quinolone nucleus was found to impart many desirable features, several derivatives of quinolone molecules were designed to bear such an atom at this position of the ring system. This had arisen a subclass of quinolone family, which is widely referred to as “fluoroquinolones” which made a revolution in the field of synthetic antibacterial agents . Fluoroquinolones possess a broader spectrum of activity, increased potency, improved oral bioavailability, enhanced pharmacokinetics, and tissue penetration and good tolerability .…”

“…Some modifications comprise ß-keto and carboxylic acid moiety that are required for hydrogen-bonding interactions with DNA gyrases or/and DNA bases, and therefore, both two functional group moieties are essential [6]. e proximity of the carboxyl and keto groups on the fluoroquinolone molecule would account for good chelating properties [7], suggesting that the interaction of the quinolone complexed to metal is a major step [8].…”

“…[3] In most of cases, it was found that Metal ions affects the action of drugs and in many cases enhance the efficacy of drugs on coordination. Therapeutically, Schiff bases and their metal complexes have been reported to exhibit a wide range of biological activities such as antibacterial including antimycobacterial, antifungal, antiviral, antimalarial, antiinflammatory, antioxidant, pesticidal, cytotoxic, enzyme inhibitory, and anticancer including DNA damage.…”
