Biologic monotherapy in the treatment of rheumatoid arthritis

Abstract: Biologics, possibly in combination with a conventional disease-modifying antirheumatic drug (DMARD) – preferably methotrexate (MTX), are used in accordance with the recommendations of the international rheumatological societies. However, in clinical practice, this recommendation is often problematic, as many rheumatologists know from personal experience. The quality of life of the patient is affected mainly by drug-induced intolerances (eg, MTX). Thus, the acceptance of the patient to treatment is often so ina… Show more

“…We did not find any difference among the three anti‐TNFs, and this was also revealed by other studies . Response to anti‐TNF treatment was higher in patients receiving concomitant MTX treatment, and this confirms, in real life, the data from therapeutic trials and registries . The male gender was sometimes a predictive factor of improved response, but this was neither the case in our study nor in Kristensen's study .…”

“…Rheumatoid arthritis (RA) is an antoimmune connective tissue disease characterized by systemic inflammation and synovial inflammation. 1 Joint synovial chronic inflammation leads to the formation of pannus, which destroys cartilage bone and surrounding tissues. Approximately 1% of the world population suffers from this disease, and it puts a tremendous economic burden on the society.…”

“…On the other hand, biological DMARDs like etanercept, adalimumab etc. showed higher efficacy, however it's use is limited due to parenteral administration, high cost and accessibility etc [3,4]. To overcome these drawbacks, Janus kinase (JAK)/signal transducer and activator of the transcription (STAT) signal pathway has been identified as one of the new therapeutic targets to treat rheumatoid arthritis.…”
