Attachment of bacterial biofilm to the surfaces of farm, fomites and equipments remains chance transmission of infection poultry and human through food chain. Formation of biofilm causes spoilage of poultry products during processing of eggs, meat and distribution. Biofilm may cause many bacterial species in biofilm society. The formation of biofilm deteriorates food quality, water supply system, drugs resistance, and reduces the efficacy of equipments, spread disease and lingering of disease course. Common bacteria cause biofilm in poultry farm and food industries are Salmonella sp., Staphylococcus spp., Listeria monocytogenes, Escherichia coli, Klebsiella pneumonae, Campylobacter jejuni, Streptococcus agalactiae. Formation of biofilm is under stress and regulated by several genes of bacterial. There are several methods of diagnosis of biofilm such as Roll plate method, tube method, microtitre assay, PCR assay, mass spectrometry method and Biological assay of Biofilm. Therapeutic elimination of biofilms for smooth production of poultry is chemical and environmental modifications. Water may be treated with several means, both chemical and physical ways. Food-contaminated biofilm-related treatment is done applying quaternary ammonium compounds, aldehydes, phenolics, alkyl amines, chlorine dioxide, etc. Veterinary medical therapy against biofilms is use of antibiotics with ultrasound, low electric current, phage therapy, nanodrug delivery system, antimicrobial peptides, antiadhesin, antimatrix and chelating substances.

Industrial food animal production plays an essential role in the global food supply chain. In parallel with the growth of the Sri Lankan poultry sector, antimicrobial usage has also been increased with the aim of reducing disease incidents. The development of antimicrobial resistance due to the irrational use of antimicrobials is a global problem. Commensals like Escherichia coli (E. coli) can easily acquire and transfer resistance to pathogenic and zoonotic bacteria which cause treatment failures in both humans and animals. The present study was conducted in 50 poultry layer (commercial chicken layers) farms in Kurunegala district of Sri Lanka during the period from November 2016 to January 2017. A questionnaire-based survey was conducted to collect information mainly on the management, biosecurity, and antimicrobial usage of selected farms. Further, E. coli were isolated from the fecal samples collected from 26 farms among those 50 farms, and their antimicrobial-resistant profiles (AMR) were investigated. Results revealed that 98% of the farms had poor biosecurity management practices while using at least one antimicrobial drug (98%). The most commonly used antimicrobial drug was enrofloxacin (79.6%) followed by amoxicillin (61.2%), both sulfamethoxazole and trimethoprim (49%), tetracycline (26.5%), neomycin (22.4%), and tylosin (4.1%). AMR profile of fecal E. coli revealed that the highest resistance is for tetracycline(81.8%) followed by nalidixic acid (54.5%), trimethoprim-sulfamethoxazole (40.9%), ampicillin (45.5%) and ciprofloxacin (31.8%). Lower levels of resistance, 13.6%, 9.1%, and 4.5% were observed for streptomycin, ceftazidime, and imipenem respectively. All the isolates were susceptible to amikacin and gentamycin; while 68.18% of isolated E. coli were multidrug-resistant (MDR). AMR and MDR findings of this study highlight the need of implementing strategies to regulate the usage of antimicrobial drugs in poultry farms in Sri Lanka, to prevent and control the emergence of antimicrobial-resistant pathogens and diseases from a ‘one health’ perspective.

Characterization of Biofilm Producing Coagulase-Negative Staphylococci Isolated from Bulk Tank Milk

Coagulase-negative staphylococci (CoNS) are considered less virulent as they do not produce a large number of toxic enzymes and toxins; however, they have been increasingly recognized as an important cause of bovine mastitis. In particular, the ability to form biofilms appears to be an important factor in CoNS pathogenicity, and it contributes more resistance to antimicrobials. The aim of this study was to investigate the pathogenic potential by assessing the biofilm-forming ability of CoNS isolated from normal bulk tank milk using the biofilm formation assay and to analyze the biofilm-associated resistance to antimicrobial agents using the disc diffusion method. One hundred and twenty-seven (78.4%) among 162 CoNS showed the ability of biofilm formation, and all species showed a significantly high ability of biofilm formation (p < 0.05). Although the prevalence of weak biofilm formers (39.1% to 80.0%) was significantly higher than that of other biofilm formers in all species (p < 0.05), the prevalence of strong biofilm formers was significantly higher in Staphylococcus haemolyticus (36.4%), Staphylococcus chromogenes (24.6%), and Staphylococcus saprophyticus (21.7%) (p < 0.05). Also, 4 (11.4%) among 35 non-biofilm formers did not harbor any biofilm-associated genes, whereas all 54 strong or moderate biofilm formers harbored 1 or more of these genes. The prevalence of MDR was significantly higher in biofilm formers (73.2%) than in non-formers (20.0%) (p < 0.05). Moreover, the distribution of MDR in strong or moderate biofilm formers was 81.5%, which was significantly higher than in weak (67.1%) and non-formers (20.0%) (p < 0.05). Our results indicated that various CoNS isolated from bulk tank milk, not from bovine with mastitis, have already showed a high ability to form biofilms, while also displaying a high prevalence of MDR.
