Bioengineered ‘golden’ indica rice cultivars with β‐carotene metabolism in the endosperm with hygromycin and mannose selection systems

Abstract: SummaryVitamin-A deficiency (VAD) is a major malnutrition problem in South Asia, where indica rice is the staple food. Indica-type rice varieties feed more than 2 billion people. Hence, we introduced a combination of transgenes using the biolistic system of transformation enabling biosynthesis of provitamin A in the endosperm of several indica rice cultivars adapted to diverse ecosystems of different countries. The rice seed-specific glutelin promoter (Gt-1 P) was used to drive the expression of phytoene synth… Show more

“…occurs within the plastids, chloroplasts and chromoplasts of fruits and flowers. 17 Their importance as light sensors for photosynthesis, preventing photo-oxidation, and as nutritional components is also well recognized, because specific carotenoids (mainly bcarotene, a-carotene, and b-cryptoxanthin) are precursors of vitamin A which have significant antioxidant activity. 18 Carotenoids constitute an important precursor reservoir for the biosynthesis of bioactive compounds in plants, bacteria, fungi and even animals.…”

“…The molecular markers were also found by crossing between transgenic and susceptible/nontransgenic varieties (Liu et al, 2009). Indica rice generations harboring the PR-3 rice chitinase gene were found tolerant to sheath blight (Datta et al, 2003). The Rir1b gene is a member of defense-related genes family.…”

“…Particle bombardment often generates large, high-copy-number transgenic loci, which are believed to be prone to instability and silencing, but there are many instances where this is not the case. For example, Golden Rice provides a clear example in which higher transgene copy numbers correspond to higher expression levels, ultimately leading to more b-carotene production in the endosperm (Datta et al, 2003). Bacillus thuringiensis (Bt)-resistant rice containing multiple transgene copies also performed well against a number of insect pests in the greenhouse (Maqbool and Christou, 1999;Maqbool et al, 2001) and under field conditions (Tu et al, 2000;Ye et al, 2001), indicating that the transgenes were expressed efficiently.…”
