Plant-inhabiting microorganisms interact directly with each other affecting disease progression.However, the role of host plant and plant habitat in shaping pathobiome composition and their implication for host susceptibility/resistance to a particular disease are currently unknown. For the elucidation of these questions, both epiphytic and endophytic bacterial communities, present in asymptomatic and symptomatic twigs from olive cultivars displaying different susceptibilities to olive knot (OK) disease, were investigated using culturing methods. OK disease was the main driver of the bacterial community, causing changes on their diversity, abundance and composition. OK disease effect was most notorious on OK-susceptible cultivar and when considering the endophytic communities. plant habitat (epiphytes vs. endophytes) also contributed to the bacterial community assembling, in particular on symptomatic twigs (knots) of OK-susceptible cultivar. In contrast, host cultivar had little effect on the bacterial community composition, but OK-symptomatic twigs (knots) revealed to be more affected by this driver. Overall, the pathobiome seems to result from an intricate interaction between the pathogen, the resident bacteria, and the plant host. Specific bacterial genera were associated to the presence or absence of OK disease in each cultivar. Their ability to trigger and/or suppress disease should be studied in the future.

Biocontrol research has the potential for managing Fusarium root rot of olive. This latter is treated by the use of isolated bacteria from the soil of olive orchards of Tunisia: Bacillus Licheniformis and Enterobacter Colcae. It was demonstrated that these two bacteria are effective against Fusarium solani and oxysporum. In an initial screening, two efficacy bacteria were isolated with a reduction of disease incidence. In vitro, both biocontrol agents were highly tolerant to the carbendazime fungicide commonly used to control Fusarium diseases. It was observed that carbendazime reduced disease symptoms at a concentration of >50 µg mL −1. The combination of the bacterial isolates and carbendazime gave a significant (P≤0, 05) control of the disease when plants were artificially inoculated with the pathogen. The application of carbendazime at a low concentration (10 µg mL −1) in combination with Bacillus B4 reduced disease symptoms by 51%, compared with a reduction of 46% obtained with the bacterium alone and no control with the chemical treatment alone. A combination of Bacillus with fungicide increased the application rate of 10 µg mL −1 carbendazime and significantly reduced disease symptoms by 74% compared to 34% with carbendazime treatment alone and, respectively, a combination fungicide and Enterobacter E4 increased the disease by 84% compared with 55% for bacteria alone. In this experiment, the integrated treatment also slightly outperformed the application of 100 µg mL −1 bacteria Bacillus and Enterobcater applied, and without fungicide also provided good disease control.

Antagonistic strainsAll antagonistic strains belong to the collection of laboratory of plant bacteriology and Biological Control at the National Institute of Agronomic Research (Meknes, Morocco). Theirs origins, isolation dates, and references are described in Tab 1.Olive knot disease caused by Pseudomonas savastanoi pv. savastanoi (PSS) is considered as the main bacterial disease of olive tree in Morocco. However, sole prophylactic measures are available. The main aim of this work was to evaluate the effects of eighteen antagonistic bacteria, as an alternative approach, to control in vitro and in planta the pathogens strains previously isolated from olive trees in Morocco. In vitro experiments revealed that many bacterial strains such as Rahnella aquatilis, Enterobacter ludwiqii, Paenibacillus brasiliensis, and Bacillus subtilis selected from collection of Regional Center of Agronomic Research, were efficient on solid media against pathogen strains. Their inhibition zones ranged from 8 to 23 mm with a higher inhibition rates found by B. subtilis strains. Enzymatic activities of antagonistic strains were assessed (cellulase, protease and amylase) as well as some plant growth promotion traits (phosphorus solubilisation and indole acetic acid production) for performance screening. B. subtilis 2515-1 was selected for pots experiment using olive trees (cv. Arbequina). This bacterium significantly reduced number and weight of knots against two pathogens strains (PSS 2064-8 and PSS 2102-4). The most reduction rates of weight and knot number were respectively 95.60% and 50%. This study suggested an alternative approach to assess the efficiency of selected bacterium in field conditions.
