Context Oxidative stress induces apoptosis within Islets of Langerhans in diabetes mellitus (DM).Enicostemma littorale blume, herb of the Gentianaceae family is used as an anti-diabetic agent across rural India. Objective This report demonstrates potent anti-apoptotic and cyto-protective activity of Enicostemma littorale MeOH extract (EL MeOH ext.) against 50 mM H 2 O 2 in isolated rat Islets. Materials and methods In this study, the whole plant methanolic extract of EL with doses 0.25-4 mg/mL each for the preincubation duration of 0.5-4 h against 50 mM H 2 O 2 were tested for optimum protective dose and time by Trypan blue dye exclusion assay. Islet intracellular reactive oxygen species (ROS) was quantified by DCFDA staining and cell death using PS/PI & FDA/PI staining. Further, comet assay, biochemical assessment of caspase-3 and antioxidant enzyme activities along with immunoblotting of PARP-1, caspase-3, TNF-a activation and p-P38 MapK (stress kinase) induction was performed. Results The optimized dose of EL MeOH ext. 2 mg/mL for 2 h was used throughout the study, which significantly decreased total Intracellular ROS and cell death. Further, caspase-3 activity, PARP-1 cleavage, p-P38 MapK (stress kinase) activation and TNF-a levels, which had been significantly elevated, were normalized. Antioxidant enzymes like catalase, superoxide dismutase, reduced glutathione and glutathione peroxidase, along with Comet assay, demonstrated that pretreatment with EL MeOH ext. can augment antioxidant enzyme activities and protect from DNA damage. Discussion and conclusions Significant anti-apoptotic and cyto-protective effects were mediated by EL with Islets of Langerhans subjected to oxidative stress-induced cell death. ARTICLE HISTORY

Background: Pharmacological treatments for diabetes are based on increasing insulin availability and improving insulin sensitivity. Today, glucagon-like peptide 1 (GLP-1) -based therapies aim to control glucose through DPP-4 inhibitors. DPP-4 is a transmembrane glycoprotein belonging to the prolyl oligopeptidase family, with the specificity of eliminating the X-Pro or X-Ala dipeptides from the N-terminal end of the polypeptides. The effect of GLP-1 in stimulating the release of glucose-dependent insulin from pancreatic islets inhibits inappropriate glucagon release after meals and slow gastric emptying promotes intestinal permeability. Study Design: The current study investigated the inhibitory activity of DPP-4 along with the antioxidant activity of Enicostemma littorale extract. Place and Duration of Study: The present study was conducted at Anurag University, Hyderabad between June-2021 to Sept-2021. Methodology: The extracts tested for a range of activities such as hydroxyl radical scavenging activity test, In-vitro DPP-IV enzyme test activity test, DPPH free radical scavenging. Results: The extract shows dose-dependent DPPH and hydrogen peroxide radical scavenging activity. In in-vitro DPP-IV enzyme assay activity, Enicostemma littorale ethyl acetate extract showed greater inhibitory activity of DPP-IV compared to vildagliptin with IC50 values of 165.64μg/ml, respectively. Vildagliptin, based on the reference standard for DPP-IV inhibitor activity, has an IC50 value of 57.44 μg/ml. According to the in-vitro analysis, the Enicostemma littorale extract has a strong inhibitory activity of DPP-IV. Conclusion: We concluded that the Enicostemma littorale will be a better source for further development as new antidiabetic drugs. To the best of our knowledge, there is no report and study on the bark part of this species.

Enicostemma littorale Blume, Syn.Enicostemma axillare (Lam.)Raynal. is a well-known plant used as drug in Ayurved, Unani-Tibbi, Siddha, Allopathy, Homeopathy, Naturopathyand Home Remedies. The present communication deals with the pharmacognostic evaluation on the different parts of Enicostemma littorale Blume (Enicostemma littorale) Family-Gentianaceae. Macroscopic and microscopic examinations of the different organ, observations and differential microchemical test have been carried out for the authentication of the samples. Physiochemical values such as the moisture contents, percentage of total ash, acid insoluble ash, acid soluble ash, water soluble ash, extractive values as petroleum ether-soluble extractives, ethanol-soluble extractives, methanol-soluble extractives and water-soluble extractives were calculated as well as color reactions of powder and extract with different chemicals were performed to observe fluorescence analysis. The extracts were subjected to qualitative screening test for various constituents. This revealed the presence protein, glycosides, alkaloids, tannins and phenolic compound, steroid reducing sugars and saponin glycosides. These observations will help in the Pharmacognostical identification and standardization of the drug in the crude form and also to differentiate the drug from its adulteration.
