Abstract: Bioassay provides a useful means of detecting and identifying plant viruses. The choice of procedure and environmental conditions for the test depends on the virus under investigation.

“…Thirty‐five of the most widely grown potato cultivars in Scotland by seed acreage were grown from pathogen‐free in vitro plantlets obtained from the SASA potato nuclear stock collection. Plants were inoculated mechanically, as previously described (Browning, ) with sap extract from PYBV isolate QV276 and PVA isolate QV282 propagated in Nicotiana benthamiana (1:5 w/v in water). Three or four potato plants per cultivar (six leaves per plant) were inoculated for each virus, and two plants per cultivar were inoculated with sap from healthy potato (cv.…”

“…Monitoring and surveillance programs must be accompanied by appropriate serological or molecular diagnostic methods [ 75 ], in addition to the characterization of the genetic diversity and population structure since novel variants can emerge. The monitoring based on symptom expression is often technically challenging, whereas different viruses induce similar symptoms, some plant species may be asymptomatic, or symptoms can even be mistaken for physiological disorders or nutrient deficiencies [ 76 ]. Serological methods, such as enzyme-linked immunosorbent assay (ELISA) and its variants, have been used for decades, allowing the identification and detection of viruses in a fast and easy manner [ 77 ].…”

“…For infection biotesting we took leaves from symptomatic plants of maize "Mosquito" and sweet maize "Brusnytsia", homogenized them with a mortar in 0.05 M potassium phosphate buffer pH 7.0 (ratio 1:10) and inoculated directly on young plant leaves (5 weeks old) with carborundum as an abrasive [42]. Three weeks after infection, the results were recorded.…”
