Bioanalytical Method for the Determination of Hydroxyproline in Mouse Kidney by High-Performance Liquid Chromatography with Tandem Mass Spectrometric Detection

Abstract: An analytical method was developed and validated for the measurement of hydroxyproline (Hyp) levels in mouse kidney by high-performance liquid chromatography with tandem mass spectrometric detection (LC/MS/MS) using an analytical column specially designed for the LC/MS/MS analysis for intact amino acids. Tissue hydrolyzed with hydrochloric acid could be directly injected into the LC/MS/MS, as well as separated and detected using the deuterium labelled Hyp as an internal standard. The calibration curve showed g… Show more

“…Thanks to the development of mass spectrometry (MS) technologies, especially coupling it to high-performance liquid chromatography (LC-MS) and tandem MS (MS/MS) in the recent years, researchers can successfully determine various analytes in different samples. [8][9][10][11] For the oxidized phospholipids, there have been reports on the determination of different kinds of oxidized phospholipids in various biological samples, including the hydroperoxides of phosphatidylcholine (PC), phosphatidylethanolamine (PE), phosphatidylinositol (PI), phosphatidylserine (PS), and cardiolipin (CL), as well as PC aldehydes (PC-CHO). [12][13][14][15][16][17][18] Nevertheless, with regard to lipidomics in diabetes, although there have been studies on lipidomics via LC-MS, 19 there are seldom reports concerning on the changes of these oxidized phospholipids.…”

“…[9][10][11][12] For example, acid chloride reagents are extremely reactive derivative reagents, which react with water, alcohols, phenols, amines, and aromatic amines. Although these reagents are generally used in a dry solvent with a base, they can also be used under alternative conditions.…”

“…1 Shiota et al developed a bioanalytical method for the determination of hydroxyproline in mouse kidney using HPLC-MS/MS. 2 A method for the determination of short-chain fatty acids in feces samples using 2-picolylamine derivatized analytes via UHPLC-MS/MS was also developed. 3 Kamao et al reported on the determination of menadione in HPLC-MS/MS using pseudo multiple reaction monitoring.…”
