Binding of the virion protein mediating alpha gene induction in herpes simplex virus 1-infected cells to its cis site requires cellular proteins.

Abstract: In herpes simplex virus 1-infected cells, the transcription of a genes, the first set of genes to be expressed, is induced by a virion component, the a-trans-induction factor, and requires a cis site. Homologs of the cis site are present in the promoter-regulatory domains of all a genes and bind two cellular proteins designated as aHl and aH2H3. We report that a-trans-induction factor, synthesized in vitro or present in nuclear extracts of infected cells, forms complexes with viral DNA fragments containing its… Show more

“…The rabbit polyclonal antibodies against VHS, VP22, or VP16 have been described elsewhere (10,28,29 2 cultures of HeLa cell monolayers were either mock-infected or exposed to 10 pfu of HSV-1(F) per cell. After 1 h, the inoculum was removed and replaced with methionine-free DMEM containing 100 Ci of [ 35 S]methionine, and the cells were incubated for an additional 18 h. PAA (300 g/ml) was present in one set of cultures throughout the infection to prevent viral DNA synthesis.…”

“…The herpes simplex virus type 1 (HSV-1) tegument protein VP16 is a structural protein of HSV-1 that activates transcription of the immediate early promoters of the virus (37,38). Although VP16 specifically activates promoters containing the so-called TAATGARAT element, specificity is conferred by the cellular DNA-binding protein(s) fused to the N-terminal domain of VP16 (39,40).…”

“…In a lytic viral replicative cycle, this DNA motif is bound by a cellular protein termed Oct-1 (Gerster and Roeder, 1988;Kristie and Roizman, 1987;. The Oct-1 DNA complex is then bound by a viral protein called the alpha transinducing factor (aTIF) or VP16 (Gerster and Roeder, 1988;McKnight et al, 1987;Preston et al, 1988). The aTIF protein is contained within infectious HSV viral particles and is therefore introduced into the cell upon viral infection.…”
