Binding of NF-Y Transcription Factor to One of the Cis-elements in the Myeloperoxidase Gene Promoter That Responds to Granulocyte Colony-stimulating Factor

Abstract: The expression of the myeloperoxidase (MPO) gene is restricted to cells of the myeloid cell lineage and is induced by granulocyte colony-stimulating factor (G-CSF). In this study, a series of deletion mutations was introduced in the promoter of the human MPO gene, which was then fused to the chloramphenicol acetyltransferase gene. The G-CSF-induced promoter activity was examined in mouse myeloid precursor FDC-P1 transformants that constitutively express the G-CSF receptor. A G-CSF-responsive element (GRE) in t… Show more

“…MPO gene expression is under the regulation of a number of transcription factors, including myeloid nuclear factors 1α and 1β 20 and granulocyte colony-stimulating factor 13 . However, there is no evidence of direct involvement of NF-κB in MPO gene regulation.…”

“…These results provide evidence of that the NF-κB signaling pathway is crucial in regulating pulmonary inflammation and MPO production. Detection of MPO activity from tumor metastases It has been shown that some malignant tumors attract neutrophil infiltration during their progression 13 . We investigated whether we could use Luminol-R to detect MPO activity in deep tissue tumor metastases.…”

“…CBF has been proved to regulate the transcription in a variety of genes, such as ferritin (Marziali et al, 1997), myeloperoxidase (Orita et al, 1997), ABO (Kominato et al, 1997), and a2 collagen, E2F1 (Hu and Maity, 2000). The function of CBF and its DNA binding site was shown by transient transfection of reporter containing the wild type and mutant CCAAT boxes, and by EMSA in which CBF bound to either wild type or mutant CCAAT sequences.…”

“…Namely, the e ects of cMyb on the transcriptional activation of GATA-1/Luc and 6MBS/Luc were quite di erent from those of DcMyb. Authentic mouse myeloperoxidase (MPO) promoter and chicken mim-1 promoter, which contain Myb binding sites and are transactivated by c-Myb (Ness et al, 1989;Orita et al, 1997), gave essentially same result (data not shown).…”
