Binding of interleukin-8 to heparan sulphate enhances cervical maturation in rabbits

Abstract: Cervical ripening is a cytokine-trigged process with substantial remodelling of the cervical extracellular matrix. Interleukin-8 (IL-8) is an important cytokine in cervical maturation. Glycosaminoglycans are also included in this process, but their role in not clearly understood. The effects of heparan sulphate (HS), hyaluronic acid (HA), IL-8, HS ⍣ IL-8 and HA ⍣ IL-8 on biochemical properties of the cervix were examined in non-pregnant rabbits. The changes in vascular pattern with collagen structure of the ce… Show more

“…In the cases where IL-8 was detected with Western blotting but no cellular staining was present in the sections, the specific band may represent the diffuse staining for IL-8 protein seen throughout the stromal tissue and in the lumen of cystic formations of the tumors. This staining may represent the binding of IL-8 to heparan sulfate, a polysaccharide present in extracellular matrix, which has been shown to enhance the biological effect of IL-8 (24,25). The high IL-8 concentrations in the malignant cyst fluids previously reported (2), together with the increasing amounts of IL-8 detected in tumor tissue with Western blotting, as observed in the present study, implies a considerable production of this protein by the tumor cells.…”

“…For example, increasing hyaluronate contributes to collagenous framework loosening and to the increase in the water content of the human uterine cervix at term 3 . In addition, hyaluronate stimulates the production of IL‐1 and ‐8, which promotes collagenase production and results in rapid cervical ripening 15,16 . Further studies of hyaluronate metabolism may help to elucidate how hyaluronate exerts its effect and how cervical ripening can be controlled during parturition.…”

“…No endotélio vascular, esta substância diminui a ação do fator de crescimento fibroblástico 24 . Em coelhos, o heparan sulfato é importante para maturação do epitélio cervical 25 e, em mulheres, há evidências de sua ação na proliferação do epitélio cervical 26 . Deve-se ainda reforçar que fatores de crescimento são mais importantes para expressão desta glicosaminoglicana do que a ação direta de estrogênio e progesterona 26 .…”
