Bidirectional transcription from a solo long terminal repeat of the retrotransposon TED: symmetrical RNA start sites.

Abstract: A single copy of the retrotransposon TED was found integrated within the DNA genome of the insect baculovirus, Autographa californica nuclear polyhedrosis virus. After excision of the element from the viral genome, a single long terminal repeat (LTR) remained behind. We have examined the effect of this solo TED LTR on the local pattern of viral transcription. Most prominent was the transcription of two sets of abundant RNAs; both originated within the LTR but extended in opposite directions into flanking viral… Show more

“…LTR retrotransposons having this same duplicate structure have been identified previously in yeast (Roeder and Fink 1983) and flies (Csink and McDonald 1995) and are postulated to be the products of homologous recombination between two full-length elements within the LTR region. (Friesen et al 1986). …”

“…protein gene (Guarino & Summers, 1986) also exhibits this conserved sequence around the transcription start site used late in infection, but not the start site used earlier. Furthermore, the two divergent transcripts directed by the solo long terminal repeat in an FP mutant (Friesen et al, 1986) begin in the middle of an imperfect palindrome whose two halves closely match the conserved sequence. Taking all these sequences together, we can discern the …”

“…Hybridization conditions were the same as for S1 mapping (a and b). The rest of the primer extension conditions were as described by Friesen et al (1986). The major primer extension signal and relevant marker sizes were identified as in (a).…”
