Beyond the Debye length in high ionic strength solution: direct protein detection with field-effect transistors (FETs) in human serum

Abstract: In this study, a new type of field-effect transistor (FET)-based biosensor is demonstrated to be able to overcome the problem of severe charge-screening effect caused by high ionic strength in solution and detect proteins in physiological environment. Antibody or aptamer-immobilized AlGaN/GaN high electron mobility transistors (HEMTs) are used to directly detect proteins, including HIV-1 RT, CEA, NT-proBNP and CRP, in 1X PBS (with 1%BSA) or human sera. The samples do not need any dilution or washing process to… Show more

“…The Debye length increases as the PBS/serum/plasma is diluted, so the surface charge carried by EGP is not screened, leading to an improved response 34 . For 0.01 × PBS, the corresponding Debye length is about 7.4 nm 35 , while the size of gold NPs is below 5 nm and the size of Ebola antibody and EGP is estimated by the vendor as 3-10 nm. As a result, the total size of gold NP plus antibody is likely within the Debye length of 0.01 × PBS, indicating the charges carried by antigen will not be completely screened.…”

“…Both curves are comparable, with a minute reduction in sensitivity. The response to HPV-16 E7 in spiked saliva decreased due to increased ion concentrations in saliva, reducing the Debye length for the biorecognition in real time [35]. The linear regime of the sensor remains and is suitable for biosensing applications, and the signal is reduced by approximately 9% compared with PBS buffer.…”

“…Since the DEP force is strongly dependent on the characteristics of the particles and the molecules bound to their surfaces, DEP has been used to isolate, concentrate, or separate different types of target particles [38][39][40][41][42][43]. However, the physics behind the frequency-dependence of the DEP force on the molecular structures is still not well understood [40,44,45].…”
